The sorter also requires at least 1 hour set up time. Book the Aria for an hour from 7am to 8am and an assistant eg: Margaret O’Prey so the machine can be QC tested and set up. You do not have to be present for the machine set up.)If the machine has not been set up in the morning you will have to add an hour to your sort request time to allow for this. SORTING REQUIREMENTS 1. 10 x 106 cells/ml. Concentrate the sample. 2. No more than 4mls of media/buffer in sterile FACS tubes. (available from stores). 3. Cell must be filtered immediately before sort (use 40um cell strainer available from stores) Note: Cells are strained and then brought to FACS room, after 20min 2 nd tube is strained and so on. This is to prevent cell clumping and hence blocking of the machine. 4. Bring tubes for collection of cells; 2 tubes per fragment collected; 2mls of media/buffer per tube. 5. Add appropriate antibiotics to sample 6. You MUST have control samples –ve and +ve 7. BE PUNCTUAL !!!!
