3.กลไกการออกฤทธิ์
Montelukast sodium is a selective and active leukotriene receptor antagonist. It binds to
cysteinyl leukotrienes (CysLT) type-1 receptors found in human airway (smooth muscle cells
and macrophages), which prevents airway edema, smooth muscle contraction and other
respiratory inflammation. The leukotrienes are also released from the nasal mucosa after
allergen exposure where montelukast sodium may inhibit symptoms of allergic rhinitis

Adult Dosing
 Asthma: 10 mg ORALLY in the evening
 Exercise-induced asthma; Prophylaxis: 10 mg ORALLY as a single dose at least 2 h
before exercise, no additional doses should be taken within 24 h of previous dose
 Perennial allergic rhinitis: 10 mg ORALLY once daily
 Seasonal allergic rhinitis: 10 mg ORALLY in the evening
Pediatric Dosing
 Asthma: (12 to 23 mo of age; granules) 4 mg ORALLY in the evening
 Asthma: (2 to 5 y; chewable tablet or granules) 4 mg ORALLY in the evening
 Asthma: (6 to 14 y; chewable tablets) 5 mg ORALLY in the evening
 Asthma: (15 y and over) 10 mg ORALLY in the evening

hypoesthesia,insomnia, myalgia, palpitation ,pancreatitis, paresthesia, pruritis, seizure,
vasculitis

hypersensitivity to montelukast sodium or any component of the product
Precausions (ข้ อควรระวัง)
1) acute asthma attack, including status asthmaticus; use not indicated for the reversal of
bronchospasm in an acute attack (Prod Info SINGULAIR(R) oral tablets, chewable tablets,
granules, 2008)
2) aspirin-sensitive asthmatic patients; use not indicated for the reversal of bronchospasm in
response to aspirin or other NSAID-related sensitivity (Prod Info SINGULAIR(R) oral tablets,
chewable tablets, granules, 2008)
3) changes in behavior and mood (including suicidal thinking or behavior and suicide) have
been reported during postmarketing experience (United States Food and Drug Administration,
2008; Prod Info SINGULAIR(R) oral tablets, chewable tablets, granules, 2008)
4) exercise-induced asthma; not indicated as monotherapy (Prod Info SINGULAIR(R) oral
tablets, chewable tablets, granules, 2008)
5) inhaled or oral corticosteroid use; do not abruptly substitute montelukast sodium for
