Investigated the induction of DNA damage by non-thermal plasma discharge in mammalian cells by looking at the phosphorylation of H2AX, a DNA damage marker. Additionally, using RNA interference, RT-PCR and molecular biology techniques, studied the ATM/ATR mediated DNA repair pathways in response to DNA damage induced by non-thermal electrical plasma produced reactive oxygen species (ROS). 2. Studied the mechanisms of Non-thermal plasma induced proliferation of endothelial cells using cell proliferation and migration assays, flow-cytometry, to develop non-thermal plasma as a potential novel therapy for growth factor initiated promotion or inhibition of endothelial cell mediated angiogenesis, which is an important target for treating diseases in which there is either poor vascularisation (chronic wounds) or abnormal vascularisation (malignancies). 3. Studied the induction of apoptosis in melanoma cancer cells by non-thermal plasma electrical discharge by using Annexin-V/PI, Caspase cleavage and flow cytometry to develop non-thermal plasma as a novel clinical device for cancer treatment. 4. Investigated the potential long term and short-term toxicity of non-thermal plasma on intact and wounded animal model to develop non-thermal plasma as a potential clinical device for wound sterilization and accelerated wound healing. 5. Studied the mechanism of blood coagulation by non-thermal atmospheric pressure dielectric barrier discharge to evaluate the use of non-thermal plasma as a potential wound healing device in a clinical or battlefield setting.
