Instructions for Use (Technical Manual content)
The Surfactant Degradation Reagent and the Surfactant Degradation Reagent for ESI
can be used to treat both solid (lyophilized or dried down) and aqueous samples
containing Progenta Acid Labile Surfactants. Both reagents are 10X concentrated
solutions.
Solid Samples
1) Preparation of solid samples: Dry down the sample containing the Progenta Acid
Labile Surfactant in a SpeedVac or lyophilizer.
2) Dilute the 10X concentrated Surfactant Degradation Reagent to 1X (e.g. add 100 µL
of 10X Surfactant Degradation Reagent to 900 µL water).
 NOTE: For samples that will be analyzed by ESI with limited desalting, the
Surfactant Degradation Reagent for ESI should be used.
3) Add 50 µL of the 1X Surfactant Degradation Reagent to the sample.
 NOTE: Add sufficient 1X Surfactant Degradation Reagent to fully dissolve the
sample.
4) Sonicate briefly (10 seconds) to mix the sample.
5) Incubate the sample at room temperature according to the times in Table 1.
 NOTE: The rate of degradation can be increased by incubating at elevated
temperatures. The degradation temperature should not exceed 55 °C.
6) The sample can be prepped for analysis by mass spectrometry, frozen for short-term
storage at +4 °C, or dried down (SpeedVac or lyophilizer) for safe long-term storage
at -20 to -80 °C.
Aqueous Samples
1) Estimate the amount of aqueous sample solution (e.g. 100 µL),
2) Add 10X concentrated Surfactant Degradation Reagent to the sample solution to a
final concentration of 1X (e.g. add ~10 µL of 10X Surfactant Degradation Reagent to
100 µL of sample solution)
3) Incubate the sample at room temperature according to the times in Table
 NOTE: The rate of degradation can be increased by incubating at elevated
temperatures. The degradation temperature should not exceed 55 °C.
4) The sample can be prepped for analysis by mass spectrometry, frozen for short-term
storage at +4 °C, or dried down (SpeedVac or lyophilizer) for safe long-term storage
at -20 to -80 °C.
