This training must be documented Safe Work MS#... Basic Task Step(s) Switch on spectrophotometer and allow to initialize. Nanodrop Hazards Electrical hazard Chemical hazard Under standard methods tab, select ‘Nucleic acid’ or protein Biological hazard Add 2ul Nuclease free water (NFW) to pedestal to initialize and click ‘OK’. After initializing machine select ‘Manual Sample ID entry’ and enter sample ID. Check the correct nucleic acid selected under sample type: either ‘DNA-50’ for DNA samples or ‘RNA-40’ for RNA samples. Add 2ul of NFW to each pedestal and click on the button ‘Blank’. Wipe pedestal clean with kimwipe. Page 2/2 Safe Procedures Wear gloves, eye protection and covered shoes at all times. Keep samples in the ice all the time. Aliquot around 10ul (typically 2ul for one detection) from each sample to avoid contamination of the samples. Prepare one eppendorf of Nuclease free water (NFW) for the purpose of cleaning the Nanodrop before and after use Bring a range of 0.5-10ul (or 0.5-2.0ul) pipettes and a rack of pipette tips Clean nanodrop with NFW properly before and after use Pipette 2 ul of sample (in duplicate) to pedestal and click on the button ‘Measure’ Once completed measurements click on the button ‘show report’ and right click on this table to export to excel.
