Thirteen isolates E(24.52%) of total number of oral streptococci was identified as Streptococcus mutans while the rest isolates were related to species Streptococcus sobrins, f Streptococcus sanguis, Streptococcus oralis, Streptococcus salivarus, and f eStreptococcus mitis which represented 18.86% ,15.09%,11.32% ,13.20%, and c16.98% respectively. t The ability of S. mutans isolates to produce slime layer was detected by using tubes method and Congo red method. The results indicated that seven isolates owhich represented 53.85% were slime layer producer by tubes method, while ten isolates which represented 76.92 % were slime layer producer by Congo red f method. G The ability of S. mutans isolates to form biofilms was detected by using microtiter plates. The results indicated that all isolates have the ability to form l ubiofilms with different thickness degree and the absorbance values at 490nm are cranged between (0.407-2.510). The minimum inhibitory concentrations of antibiotics (Ampicillin, Amoxicillin, Cephalexin and Erythromycin) to S. mutans isolates had been determined. They were 1 µg/ml, 32 µg/ml, 16 µg/ml, and 64 µg/ml respectively for the isolate S1 and they were 1 µg/ml, 64 µg/ml, 32 µg/ml, and 64 µg/ml respectively for the isolate S7. The minimum inhibitory concentrations of chemical inhibitors (Crest, Brushup, Stannous fluoride and Zak) to S. mutans isolates had been determined. They were 0.01mg/ml, 0.023 mg/ml, 0.0125 mg/ml, and 0.0037 mg/ml respectively for isolate S1, and they were 0.02 mg/ml, 0.023 mg/ml, 0.025 mg/ml, and 0.0075 mg/ml respectively for isolate S7. The effect of sub minimum inhibitory concentrations of antibiotics (Ampicillin, Amoxicillin,Cephalexin,and Erythromycin)on biofilms formation by S1 and S7 isolates in microtiter plate had been tested .The results indicated that these concentrations of antibiotics inhibited bacterial adhesion on polysteren surface of microtiter plates and consequently caused biofilms detachment and this causes a decrease in absorbance values of biofilms. The effect of sub minimum inhibitory concentrations of chemical inhibitors (Crest, Brush-up,Stannous fluoride and Zak)on biofilms formation by S1 and S7 isolates in microtiter plate had been tested . The results indicated that the values of biofilms decreased after treatment with sub minimum inhibitory concentrations of chemical inhibitors. Glucan binding proteins (GBPs) were extracted and purified by using affinity chromatography on Sephadex G-150 column with dimension of (1.2 x 10) cm and with flow rate (30ml/hr). The glucan binding proteins were eluted from Sephadex G-150 by (3M) guanidine HCl. Antibodies (Anti-GBPs) were prepared for this protein by injection this protein in experimental animal. The titration of anti-glucan binding protein (antibodies) for glucan binding protein of S. mutans in serum of rabbits (control and experimental) was determined by using passive haemagglutination test. The effect of anti-GBP on biofilm formation by S. mutans was tested by using microtiter plates. The results indicated that absorbance values of biofilm decrease. Also results showed significant effect on biofilm formation after treatment with dilution of anti-GBP. Experimental immunization with S. mutans antigens involved in the adherence and accumulation of bacteria to tooth surface (Gbp) have been shown to induce antibody response.
