Ciliopathies are a class of human developmental conditions that are of increasing relevance to basic research and clinical practice. The post is funded by a Medical Research Council project grant and will contribute to the execution, analysis and interpretation of siRNA-based reverse genetics screens for novel ciliopathy genes and processes. The siRNA screens will be performed in collaboration with the BioScreening Technology Group (BSTG) at the University of Leeds. The project will require an excellent knowledge of modern techniques of genetics (including next generation sequencing methodologies) and advanced imaging techniques (including confocal microscopy and high-content imaging). You will also evaluate screen hits using functional assays, including the assessment of protein-protein interactions using affinity purification methods (TAP-tagging), so previous experience of these systems is desirable. A good knowledge of biochemical assays is essential. Candidates must be enthusiastic, highly motivated and demonstrate a high level of relevant laboratory experience and technical competence. In addition, the post holder must show the ability to manage their time effectively and work under pressure, exhibiting effective interpersonal, communication, and team-working skills. The project will involve cell culture, including derivation of mouse embryonic fibroblasts, advanced microscopy techniques, recombinant DNA technology, and will have a significant component of bioinformatic and image analysis. BioScreening Technology Group (BSTG) The BioScreening Technology Group (BSTG) provides an interactive service to enable the characterization of the function and interactions of particular proteins, and support the identification and validation of potential biomarkers (diagnostic, prognostic or predictive) and therapeutic targets. More specifically, the Group helps users to develop and formulate plans for the execution of projects to: (1) develop probes for biomarkers/drug targets and (2) undertake reverse genetic screens. The BSTG is funded by the Leeds Biomedical and Health Research Centre to promote collaboration between basic sciences, translational and clinical research. The reverse genetics arm of the service consist of a manager, postdoctoral fellow and a technician who are all based in the section of Ophthalmology and Neurosciences.
