Flow Cytometry and Cell Sorting Facility

Guidelines for obtaining access to facilities and services

There are three main services provided to researchers by the facility:

1.

A sample analysis service whereby researchers submit appropriately prepared samples to experienced facility staff for analysis.

2.

Training to enable researchers to use the flow cytometer analysers for their studies.

3.

Cell sorting applications (performed by facility staff only).

Sample analysis service.

If you do not intend to perform flow cytometric analyses on a regular basis, or do not have the time to invest in learning to use the technology and analyse the data, you may prefer to allow us to analyse your samples for you. Alternatively you may wish us develop a new assay for you. When using this service the cost will include instrument time, operator time and the analysis of data (provided in a standard format).

At present we have a number of standard analyses that we can perform for you, the cost of which is easy to predict (see below). More complex analyses (e.g. immunophenotyping) or assay development vary greatly in complexity and therefore the data analysis time can vary greatly. This makes it difficult to cost this type of experiment but we can advise on the likely cost before they are performed.

To make use of this service:

 If planning a new study, arrange to meet with the relevant facility staff at least a week before staring the experiment so that we can discuss experimental design, necessary controls and sample preparation.

 Prior to accepting a booking we will need a cost-centre number to which charges will be made.

 Book a date and time for the analysis to be performed. We can get very busy so please give at least a week notice when making a booking and be prepared to fit in around bookings that have already been made.

 You will be provided with a detailed protocol for the preparation of samples suitable for flow cytometric analysis. It is essential that this protocol is followed correctly, and that the correct number of cells and suitable controls are provided for analysis.

 Incorrectly prepared samples will only be processed at our discretion and you will still be charged for our time. Incorrect cell numbers or samples containing clumps can considerably increase the time it takes to analyse samples, and therefore the cost.

 Please be punctual bringing samples for analysis. You will be charged for any time we are waiting for you.

Training in the use of flow cytometers within the facility.

As everyone is different and researchers have different requirements, we like to tailor all training to the individual. Therefore training tends to be on a one-to-one basis. Training is provided on the use of the instrumentation, acquisition of data, and analysis of data produced. Flow cytometry is a complex technology and can be used in a variety of different ways. Proficiency in one particular technique does not signify the ability to perform other techniques, so please seek advice when trying something new. We are always here to help in any way, as it is important to us that users make the most of the technology available and use it to produce meaningful results.

To access this service:

 We can be very busy at times so you will need to give us a minimum of a week notice when requesting training.

 Only request training when you are able to use and practice the techniques you have learnt on a regular basis.

 Arrange to meet with us to discuss your experiments: We may suggest better alternatives or improvements and will determine which instrument is most suitable for your needs.

 We will arrange a date for the initial training session introducing you to the use of the cytometer. Before this session you will need to have achieved a basic understanding of the principles of flow cytometry.

 Prior to being able to access the booing system we will need a cost-centre number to which charges will be made.

 We will then give you access to the Bumblebee on-line booking system.

 After the initial training session we will arrange a further session when we can assist you running samples yourself.

 Additional assisted sessions will be arranged until we, and you, feel comfortable going it alone.

 The instrument booking system is used to determine instrument usage and therefore charges. Please be aware that all booked time will be charged.

Cell sorting.

Cell sorting is a complex technique and to do it correctly requires considerable training.

Therefore all cell sorting applications in CLS are performed by facility staff. It is very important to us that you get what you want in the most efficient way possible. We don’t want to waste time sorting samples unnecessarily any more than you want to pay for it! We can only ensure this by planning the sort carefully in advance.

 If you wish to perform a sort that you have not performed before you must arrange to meet with us well in advance so that we can discuss technical issues.

 We will need detailed information to be able to determine whether it is possible to get what you need.

 We will calculate how much sample you will need to provide and how long it will take to take to collect the required number of cells.

 We will then book a mutually convenient time for the sort to be performed.

 Sorting appointments can fill up very quickly so please give us at least one weeks notice when booking a sort. We may be able to accommodate last minute bookings but do not expect this to be the norm.

 For a successful sort it is critical that samples are prepared correctly and in accordance with our instructions.

 Please be punctual bringing samples for sorting. You will be charged for any time we are waiting for you and we will not extend the sorting time to accommodate a late arrival.
