Full Biology, Environmental Studies, Science Education Full The primary focus of my research is the regulation of cancer progression and metastasis in non-small cell lung cancer. Specifically, I study the regulation of plasma membrane transport protein, the Na-H Exchanger Isoform 1 (NHE1) and the multiple roles it plays in supporting cancer development and progression. NHE1 is a key effector protein involved in the coordination of intracellular events and extracellular microenvironment supporting neoplastic transformation. Activation of NHE1 is one of the initial steps in the development of the transformed phenotype of cancer cells. This activation leads to an alkalization of intracellular pH (pHi), which is a permissive requirement for cell growth. During tumor cell migration, NHE1 localizes to the leading edge of cells where a combination of an elevated pHi and the binding of a range of proteins to NHE1 stimulates the formation of actin filaments that drive the forward movement of these migrating cells. To facilitate migration and invasion, NHE1 activity acidifies the extracellular pH (pHe) at the leading edge of migrating cell stimulates the activation of a range of extracellular proteases that digest the extracellular matrix (ECM). Using both lung cancer cell and mouse models, we have shown that inhibition of NHE1 significantly reduces cancer related behavior including tumor invasion and metastasis and morbidity.
