CHAPTER-III Method of Study For selection of Cholesterol Affected individual person Advertise published in news paper that person who have cholesterol problem if they want free treatment on their disease without taking drugs, only cholesterol control by exercise and food supplement can call on this mobile No. 7709003653 for taking part into cholesterol controlling programme. 150 person Men and women examined and declared 100 of them were medically fit for this study and they were selected by lot method and they divided randomly in to two groups, Men and women group separated as one control and one experimental group. The selected Hundred subjects were randomly divided in to four groups of twenty five each, out of which group I (N-25) Underwent running activity and Food Package and group II (N-25) remained as control. Pre test were conducted for all two groups on Low Density Lipoproteins, Triglyceride, Total Cholesterol and High Density Lipoproteins. The experimental group participated in their respective Exercise programme including Walking and other Aerobic for a period of Twelve month. Post test were conducted on the above mentioned dependent variables after Twelve month of the training period. The training programme scheduled at morning 6.00 a.m. to 7.00 a.m. and evening 6.00 p.m. to 7.00 p.m. The advises were given walking exercise for 30 minutes at morning and evening. Every exercise session should include a warm-up for 5 to 10 min. must mean we have to stress your hand and legs relax, neck by moving roundly then start exercise, at the time of exercise pulse rate must be increase 45% to 80% then functional capacity and then stay constant for 60 min. at the time of stopping exercise, stop slowly for 5-10 min. cool down phase. 17 During this time they were closely observed, their dietary pattern assessed by a questionnaire. They were given knowledge about the risk factors of coronary artery disease and that disease could be reversed by changing diet. they were advised to follow a modified diet schedule based on Diet Manual. The modified diet included more vegetables and fruits, with moderate amount of rice and pulses. They were advised to keep away from the possible risk factors like alcohol, smoking. Cooking Food Package:Before exercise: 1 Cup green tea or skimmed milk,/ Fenugreek seed + wheat Bran + semolina make sweetmeat ball of normal size Breakfast: 50gm of cereal + 25 gm of leg tunes + 250gm of Vegetables (like salad or curries). Or 200ml Any Juice – Orange, pineapple, Sweet lime, carrot, luffa Lunch : 50gm. of cereals + 50 gm. of leafy vegetables + 1 00gm. of other vegetables. 4 O ‘clock : 1 glass of any type of juice + any types of fruits 1oo gm. Dinner: 2 Chappathees with vegetable cury or equivalent. Or Khichari like jawar, maize, pigeon pea. Oil – Ground nut, karadi, sesame (Patients were supplied with a dietary guide describing various choices of food). If patients like to take non vegetarian food, they were allowed to take 100 g of fish thrice in a week, 100 g of skin removed chicken twice in a month. They were also allowed to take egg white, but not egg yolks. Sources of data To execute these investigation twenty five males and twenty five females of available abnormal Cholesterol person having age 45 year or more who did lipid profile test and angiography, Who having no blockage in main blood vessels and do 18 not have tendency to do bypass surgery and angioplasty on him or obese person. Lazy whose life style is Luxury or without hard work, diabetic person one whose have heredity of heart disease Cholesterol person selected in Nagpur municipal corporation only. 25 men and 25 women applying controlling program of selected abnormal cholesterol person and after one year conduct lipid profile test and data collected then necessary statistical techniques applied to find the To study Effect of Selected Exercise Module and Food Package on Cholesterol Level of Affected Individuals. The research scholar use mean, standard deviation and t-test for interpreting the results. Criterion Measure and Administration of Test For determination of the Cholesterol level or lipid profile test, estimate with the CHOD/PAP ( CH- Cholesterol, OD- Oxides, P- Phenol, A- Amino antipyrine and P-Peroxide ) most popular color metric methods The blood sample of the subject taken before 10 to12 hours of fasting in the morning session. 1) The blood sample of the subject collected with the help of clinical laboratory technician by using disposable syringe. 2) 2 ml blood sample collected from the subject 3) Blood sample taken in 5 ml test tube and allow to clot blood sample. 4) The blood sample centrifuged for 5 min till the serum from R.B.C. are separated 5) Then blood samples stored at 2.8 c and the level of Cholesterol estimated in pathological laboratory. 6) Estimating total Cholesterol level i) 1000 μml Cholesterol reagent taken and add 10 μml serum sample to it. 19 ii) Take 1000 μml Cholesterol reagent and add 10 μml Cholesterol standard in to test tube iii) It incubated it at 37 c for 10 min. iv) After incubation taken OD (Optimal Density) of test and standard against reagent blank. v) Total Cholesterol calculated through following formula Abs. of test Total Cholesterol in Mg/DL = Abs of Standard x 200 7) Estimating of Triglyceride level i. 1000 μml Triglyceride reagent taken and add 10 μml serum sample to it. ii. Take 1000 μml Triglyceride reagent and add 10 μml Triglyceride standard in to test tube iii. It incubated it at 37 c for 10 min. iv. After incubation taken OD (Optimal Density) of test and standard against reagent blank. Total Triglyceride calculated through following formula Abs. of test Triglyceride in Mg/DL = Abs of Standard x 300 8) Estimating of HDL level Step –ii i. 1000 μml HDL reagent taken and add 50 μml serum sample to it. ii. Mixed well and incubate at room temperature for 5 minute. iii. The above tube centrifuged at 2500-3000 rpm. From centrifuge machine 20 iv. To all clear supernatant form above solution. Step –ii i. Take 1000 μml HDL reagent and add 50 μml Supernatant from step-i ii. Take 1000 μml HDL reagent and add 50 μml Standards in to test-tube iii. It incubated it at 37 c for 10 min. iv. After incubation taken OD (Optimal Density) of test and standard against reagent blank. HDL calculated through following formula Abs. of test HDL Cholesterol in Mg/Dl = Abs of Standard x (25 x 2) 9) Estimating of LDL level LDL Cholesterol level calculated with the help of total cholesterol, Triglyceride and HDL through following formula. LDL Cholesterol in Mg/Dl = Total cholesterol-{ Triglyceride 5 } - (25x 2) 11 Then estimating of VLDL level With the help of LDL level through following formula LDL Cholesterol in Mg/Dl = Triglyceride 5 12. The estimated total cholesterol, Triglycerides, LDL VLDL, HDL Cholesterol value compare with normal reference values as fallow. 21 Types of Cholesterol Values Category Total Cholesterol Less than 200 Desirable Mg/dl LDL Cholesterol 200-239 Borderline 240 and above High Less than 100 Optimal Mg/dl HDL Cholesterol Triglycerides 100-129 Near Optimal/ above optimal 130-159 Borderline High 160-189 High 190 and above Very High Less than 40 Low ( High Risk ) 60 or highest High ( Optimal) Less than 150 Optimal Goal
