Figure 1
Proliferation assay in MiaPaCa-2(A) and Panc1(B). There was no difference of growth curve
between Ep-CAM transfected cell and the others. Ep-CAM; Ep-CAM expression vector
transfected cell line, Mock; Mock vector transfected cell, Control; the parental cell as
control.
Figure 2
The pictures of invasion and migration assay in MiaPaCa-2 and Panc1. Invaded or
migrated cells through the membrane were stained. There were apparently less invaded or
migrated cells in Ep-CAM transfected cell line than the others (Bar=50µm). Ep-CAM; EpCAM expression vector transfected cell line, Mock; Mock vector transfected cell, Control;
the parental cell as control.
Figure 3
Relationship between Ep-CAM expression and overall survival rate in each institute. Both
in Osaka Univ. and OMCC, Ep-CAM high expression group showed significantly good
prognosis (3-year survival; 55.4% versus 19.7%, p=0.016, 58.7% versus 17.8%, p=0.048,
respectively).
Figure 4
Reanalysis of overall survival by dividing all patients into two groups, LN negative
group(n=40) and LN positive group (n=55). LN negative group showed bigger difference of
prognosis between Ep-CAM high and low group than LN positive group.
Figure 5
The comparison of EMT related gene expression, CK19 (A), α-SMA (B), and Fibronectin
(C), between Ep-CAM transfected cell lines and non-transfected cell lines. There was no
significant difference among each group.
Figure 6
The relationship of EMT related protein expression, CK19 (A) and α-SMA (B), to Ep-CAM
scoring. There was no significant relationship among each group. Immunostaining for
fibronectin was not performed because we couldn’t get a good antibody of fibronectin for IHC.
Figure 7
We revaluated the relationship between Ep-CAM expression and overall survival by
dividing enrolled 95 patients into three groups, “high expression” with over 6 of scoring
(n=20), “middle expression” with 3 to 6(n=55), and “low expression” with under 3 (n=20).
High expression group showed better prognosis than the other two groups and the overall
survival rate decreased gradually according to the level of Ep-CAM expression with a
marginally significance (p=0.09).
