SUMMARY OF RESEARCH INTERESTS
Prof. Alcalay has been actively involved in the study of the molecular defects underlying acute myeloid leukemia (AML) since
1987. Major achievements in this area include: cloning of the 15;17 translocation characteristic of acute promyelocytic
leukemia, functional analysis of the PML/RAR fusion protein, analysis of transcriptional regulation associated to expression of
the leukemogenic fusion proteins PML/RAR, PLZF/RAR and AML1/ETO, identification of mutations in the nucleophosmin
(NPM) gene in AML patients with normal karyotype, identification of a specific expression profile associated to AML with NPM
mutations, analysis of AML1/ETO genomic binding pattern and identification of the cell-cycle inhibitor p21 as a crucial player
in the maintenance of self-renewal of leukemia stem cells. She has solid experience in the use of functional and genomic
approaches, and has focused much of her interest in the past decade on exploiting an integrated genomic approach to study
the molecular basis of acute leukemias.
Her current interests include analyses of transcriptional networks underlying normal hematopoietic differentiation and their
subversion in the pathogenesis of leukemogenesis through the integration of data deriving from mRNA and miRNA
expression profiling, chromatin immunoprecipitation coupled to deep sequencing (ChIP-seq), identification of histone
modifications and proteomic analysis of transcriptional complexes containing leukemia-associated oncoproteins. Other
projects addressing the discovery of somatic mutations with prognostic/predictive relevance in AML, and oncogene-induced
activation of stem-cell signalling pathways in leukemia are ongoing in her laboratory.
Prof. Alcalay has always supported her independent research through funding obtained as Unit Leader from the Italian
Association for Cancer Research (AIRC), the Ministry of University and Research (MIUR), the Italian Ministry of Health, the
Cariplo Foundation and the Umberto Veronesi Foundation (FUV).
