How to run a PT test A/ Reagent Preparation • 🥤 Transfer the entire contents of one vial of Solvent (R2) into one vial of Néoplastine® (R1) of the same kit. • 🕕 Allow the reconstituted reagent to stand at room temperature (18-25°C) for 30 minutes. • âž° Swirl the R1 vial gently to obtain a homogeneous suspension. B/ Analyser Preparation • • While the analyser is off, hold down then push Here you can set the ISI and MNPT values, using buttons 2, 3, and 4 as indicated on the screen • Then you can switch the analyser on • Then wait for it to reach 37° The reconstituted Néoplastine® needs to be incubated in the 37°C reagent spot on the analyser, not filled higher than the top of the space • Place cuvettes in the cuvette incubation locations, with a metal ball in each cuvette for at least 3 minutes to heat the ball up • Then you need to choose the incubation time for the PT test to 120s by Holding down then pushing until the desired incubation time is Visible Director: K N Dodkins C/ Test Procedure • 🔥 In an appropriate cuvette with ball at 37 °C, pipette 100μl plasma (standard, patient’s or control) at the 12o’clock position in the cuvette, choose the test by pushing and then to choose the PT test which will start to incubate the sample for 120 seconds at 37°C. • When there is only 20 seconds left of the incubation, the analyser will indicate this every 5 seconds by beeping once, move the cuvette to the measurement well • A longer tone indicates the end of the incubation, then a “start” appears on the screen. You have 5 seconds to active the measurement mode – activate the measurement mode by pushing the button • 🥤 Add 200μl of pre-warmed Néoplastine® at the 6 o’clock position and simultaneously start measurement for clot formation by pushing the button. The pipetting of the pre-warmed Néoplastine® at the exact same time as you push the button is very important, as this indicates the start of the clotting time • Record the time, in seconds (T)/Ratio (R)/INR (I), this flashes on the screen D/ Stability Once reconstituted, R1 remains stable for: Stability: 8 hours at 37 °C Stability at room temperature: 48 hours at 20 ± 5 °C Stability in fridge: 8 days at 2-8 °C.
