The CycloGraph™ System is a centrifugally accelerated
device for performing preparative chromatographic
separations. It combines the advantages of both
preparative TLC and column chromatography and delivers
fast, efficient separations - usually in 20 minutes or less.
To obtain a separation, a solution of the sample is
applied inside the adsorbent ring of the pre-cast rotor
using a pump or hand syringe. The solvent mixture
is then pumped through the adsorbent layer as the
rotor spins. Centrifugal force separates the individual
components because of their different affinities for
the layer and the solvent mixture. As the individual
components/rings reach the outer rim of the rotor, they
are spun off and collected in a special circular trough for
retrieval and further testing.
Typical flow rates are in the range of 2-3mL/min
per millimeter of adsorbent thickness. The inlet of the
solvent metering pump may be switched from a weak
solvent to a more polar solvent during a run. This
allows compounds with a wide range of polarities to be
separated. Use of a strong solvent at the end of a run
removes strongly adsorbed compounds and allows reuse
of the rotor. Use the built-in UV254 lamp to monitor
the separation as it occurs. The separated substances
which adsorb 254nm light will be visible against the
fluorescent green background of the rotor.
CycloGraph™ II – Universal System
This unit operates using 100-240 V at 50 or 60Hz.
The CycloGraph™ is CE marked and supplied with a US
cord set, but also has an IEC 320 compatible outlet
for universal operation. (Note: IEC 320 cord set is not
supplied with the unit.)
The CycloGraph™ II System
includes the instrument, Lowflow (0-15mL) Solvent Pump with
Tubing, Rotor Scraper Kit, Built-in
4-watt UV254 UV Lamp, Dropin rotor mount for easy set-up,
instruction manual and one year
warranty.
Other features of the unit are
the convenient hinged lid with
safety interlock, fixed speed motor, touch pad control
switching system and an enlarged tempered glass view
port.

The scoring and cutting of
glass plates is routine in many
chromatography labs, either to economize on plate
usage or to cut the plate following separation for further
analysis.
This Basic TLC Laboratory
Starter Kit contains all of the
supplies necessary for the
least experienced student or technician to complete an
analysis using basic TLC techniques.
The TLC Plate Cutter has a high quality carbide scriber
mounted into a movable plastic head. It is designed to
produce optimal scoring on the glass surface of 20x20cm
TLC plates with a total thickness (glass + coating) of
2mm. A smaller template is included with the unit for
ease in cutting 10x10cm plates. The carbide scriber is
easily replaced when there is a noticeable deterioration
in cutting efficiency.
The kit includes a 20x20cm glass development
tank and lid, tank liners, Silica Gel G TLC plates,
sample spotters, a reagent sprayer, a disposable spray
box, spotting template, sample dye mix and a basic
instruction manual.

Note About Sample Applications
Thorough drying of samples is a step
that is often ignored but can result
in development errors. If a small
amount of solute is left on the plate
it can interact with the development
system and affect the separation.
Many analysts assume the plate is dry when
the solvent is no longer visible on the plate (5-10
minutes). However, depending on the volatility of
the solvent and the quantity of sample applied, the
drying time can be considerably longer. Generally,
Rƒ values decrease as drying time increases until the
dry state (stable Rƒ) is reached.
Plates can absorb moisture from the atmosphere
during storage, so prior to sample application, all
TLC plates should be heated at 120-130°C for about
30-40 minutes and allowed to cool in a desiccator.
This ensures reproducible Rƒ values.

The solvents used for the separations are common
laboratory reagents and are not provided in the kit.
Also, due to transportation regulations, a spray reagent
for analysis of samples other than the dye mix must be
purchased separately.

The TLC Sample Streaker
is designed to apply a narrow
band (1mm wide) of sample
solution to a plate up to
40cm wide. Without scratching the adsorbent layer, the
streaker will apply sample in one-tenth the time required
for hand spotting.

For an effective separation, the solution must be
applied to the plate in a continuous band, not separate
spots as in analytical TLC. This is accomplished through
the mechanical action of forcing a syringe plunger
downward as it travels across a sloping stainless steel
bar. Complete instructions for use and calibration of this
device are included.

The Spectroline® CC-80 cabinet holds two 20 x 20cm
TLC Plates. Bottom panels of the cabinet are removable
and allow placement on top of the transilluminator. The
cabinet is constructed of aluminum with a chemical
resistant polyurethane coating. Felt curtains block
out light. An internal white light provides visible

For UV Photo Documentation – Digital Camera with
Darkroom
For UV photo documentation, a developed TLC Plate is
placed in the darkroom cabinet and flooded from above
with the appropriate UV light. The flexible eyepiece
is replaced by the snap-on cabinet adapter provided
with the camera. The camera is placed on the adapter
and connected to the lab computer via the USB port. A
simple click records the image.
Using the visible light method with mounting stand
to record the plate image, the camera is placed on the
holder which is preset at the appropriate focal distance.
A simple click records the image. The camera mounting
stand is especially helpful in archiving old photos of
plates because it allows adjustment of the focal length
using the height adjustment of the stand. The camera
stand provides a better light source for regular visible
light photos.
The software program provided with the camera allows
easy archiving and retrieval of the photo images. The
USB port connection from the camera to the computer
makes use of the documentation system fast and easy.
The documentation process is very simple but provides
long term, accessible data storage.

In some cases it may be helpful to use a transilluminator to enhance the visibility of the
fluorescent compounds. The bottom panel of the darkroom cabinet can be removed and the entire
cabinet can be placed on top of the transilluminator surface. The Transilluminator, 365nm, allows
the long-wave light to pass through the TLC plate and enhances the visibility of the fluorescent
compounds. While optional for most TLC work, it can be helpful when additional contrast is needed.
