You have chosen a new, innovative technology for measuring oxygen and pH.

Chemical optical oxygen and pH sensors (also called optodes) have several important features:

They are small.

Their signal does not depend on the flow rate of the sample.

They can be physically divided from the measuring system which allows a noninvasive measurement.

They can be used in disposables.

Therefore, they are ideally suited for the examination of small sample volumes, for highly parallelized measurements in disposables, and for biotechnological applications.

A set of different oxygen and pH minisensors, flow-through cells and integrated sensor systems is available to make sure you have the sensor which matches your application.

2 Description of the T-Flask Adapter

The T-Flask Adapter is used for cell culture flasks with integrated sensors to conduct culture monitoring with the SFR Shake Flask Reader. It is designed to guarantee optimal positioning of the vessel and alignment above the optical module of the SFR for sensor read-out. The respective T-Flasks with integrated sensors are available in different types and sizes, and are all treated so they fit on the adapter. They come pre-calibrated and irradiated, and are readyto-use. Up to 9 T-Flask Adapter can be attached on top of the SFR. The T-Flask Adapters can also be used in combination with adapters for other cultivation vessels, e. g. clamps for shake flasks, or the iTube Adapter for cultivation tubes.

Up to 9 T-Flask Adapter can be attached on top of the SFR. You need a personal computer with installed SFRS Shake Flask Reader software and installed Bluetooth adapter (SFRS and

Bluetooth adapter are delivered with the SFR Shake Flask Reader). The T-Flask Adapters can also be used in combination with adapters for other cultivation vessels, e. g. clamps for shake flasks, or the iTube Adapter for cultivation tubes.

3.1 Mounting the T-Flask Adapter on the SFR

Put the T-Flask Adapter on the SFR so the optical module is placed under the opening. Then slide the adapter into place so the adapter´s holes match the holes on the SFR (see Fig. 3).

Place the four screws in the holes like shown in Fig. 4. Press down the T-Flask adapter and tighten all four screws. Make sure the adapter is attached to the SFR firmly so it does not come off during measurements.

3.2 Possible Adapter and Flask Positions on the SFR

Operation

You can place up to 9 T-Flasks with integrated sensors on the SFR. The T-Flask Adapters have to be attached according to Fig. 5 with two rows facing in the same direction and one row facing in the opposite direction.

3.3 Positioning of the Flask on the Adapter

Operation

Now you can place the T-flask equipped with sensor spots and containing your media in the adapter. There are four knobs on the bottom of each T-flask with sensors and four recesses in each T-Flask Adapter to ensure the correct position of the flask and its integrated sensor spots. Put the T-flask on the adapter and slide it so the four knobs snap in the respective recesses on the adapter (see Fig. 6). As there are two bigger and two smaller knobs on the flask it is easy to determine which way the T-flask has to face, as the respective recesses on the adapter are also different in size. This way the sensors are aligned correctly for read-out with the SFR.

In case you want to shake the flasks, please note: The maximum shaking speed is 60 rpm!

3.4 Measurement

Slide the T-flask in position, so the knobs snap in the respective recesses on the adapter

Calibration

The T-flasks are delivered irradiated and pre-calibrated. Please download the calibration file(s) matching the batch number on the flask(s). You can find these files on www.presens.de/calibration . Then load the respective file(s) with the SFR software.

Equilibration

The sensors need to be equilibrated before usage. In order to do so you have to fill the flasks with your media and wait for at least 3 hours so that the sensors can equilibrate.

One-point adjustment pH one-point adjustment is advised if your media differs from physiological conditions.

(Please see the SFRS instruction manual for more detailed information on one point adjustment.)

Re-calibration

If you have used the pH sensor for more than 10,000 measurements, a new one-point adjustment is mandatory to ensure the accuracy of your measurements. The oxygen sensor requires no re-calibration up to 100,000 measurement points.

Do not use the T-flasks with integrated sensors for more than 100,000 measurement points.

Operational notes during measurement

The SFRS is for controlling your measurement. It offers different options to display and monitor your measurement data:

1. Virtual tray: showing the DO and pH values of all 9 positions

2. 9 positions graph: showing the graph of DO and pH of all 9 positions

3. Detailed graph: detailed graph of one position

4. Cumulated graph: for overlaying different graphs of the positions

First you should check whether an error or “no sensor detected” message is displayed for pH or DO measurement (see Fig. 7). If this is the case, the flasks might not have snapped in properly.

Please check, if you have placed the T-Flask Adapters and/or flasks in the right way and adjust their positioning if necessary. Check the signal quality of the sensors again. If the problem persists, please contact our service team.
