The optical bench of the MACSQuant
Analyzer has three lasers (violet, blue
and red). The three lasers are spatially
separated and are focused on the
cuvette in three different locations.
As the cells flow through the cuvette
one at a time, light is scattered and
collected at 180° (FSC) or is deflected
through an objective at 90° angle (SSC
and fluorescence signals). The SSC and
fluorescence signals are separated by
various filters and are collected in the
appropriate photomultiplier tubes (PMTs).
