The three lasers are spatially separated and are focused on the cuvette in three different locations. As the cells flow through the cuvette one at a time, light is scattered and collected at 180° (FSC) or is deflected through an objective at 90° angle (SSC and fluorescence signals). The SSC and fluorescence signals are separated by various filters and are collected in the appropriate photomultiplier tubes (PMTs).
