Cat Nr/REF: KB-10107 Edition 19.05.06 English For professional use only Poseidon DLEU (13q14) & p53 (17p13) Probe Introduction: Deletion at 13q involving the band q14 occurs frequently in B-cell chronic lymphocytic leukaemia (CLL) and Multiple Myeloma. A minimal critical region has been shown to lie between the RB1 gene and the marker D13S25 containing DLEU1, DLEU2, and RFP2 genes. Alterations of the p53 gene (TP53, 17p13) occur not only as somatic mutations in human malignancies, but also as germline mutations in some cancer-prone families with LiFraumeni syndrome. Deletions of p53 are frequent in CLL and MM, usually associated with unfavorable prognosis. Intended use: KB-10107 ON DLEU (13q14) / p53 (17p13) The DLEU (13q14) specific DNA Probe is optimized to detect copy numbers of the DLEU gene region at 13q14. The p53 (17p13) specific DNA region is optimized to detect copy numbers of the p53 gene region at 17p13. The probe is recommended to be used in combination with a Poseidon FISH Kit providing necessary reagents to perform FISH (KB-60002, KB-60003 or KB-60001) for optimal results. Critical region 1 (red): The DLEU (13q14) specific DNA probe is direct-labeled with PlatinumBright550. Critical region 2 (green): The p53 (17p13) specific DNA probe is direct-labeled with PlatinumBright495. Reagent: Poseidon probes are direct-labelled DNA probes provided in a concentrated format. For FISH this probe needs to be diluted with the supplied hybridization buffer. For hybridization use 2 parts of probe + 8 parts of Cell Hybridization Buffer (CHB). Please refer to the Instructions for Use for the entire Poseidon FISH protocol. Note: For tissue hybridization use THB (Tissue Hybridization Buffer) and KREABoost – available on request. Deletions involving the DLEU gene region will show one red signal and two green signals for the p53 region at 17p13 (1R2G). Deletions involving the p53 gene region will show one green signal and two red signals for the DLEU region at 13q14 (2R1G). Deletions involving both critical regions at 13q14 and 17p13 will show one red and one green signal only (1R1G). Two single colour red (R) and green (G) signals will identify the normal chromosomes 17 (2R2G).
