polymerase from your PCR sample while providing high recovery of DNA fragments ranging in length

The Diffinity RapidTip2 functional pipette tip contains
everything you need for PCR purification. The tip
is filled with our proprietary adsorption technology
that has a differential affinity for PCR reaction
components. The impurities (e.g., single-stranded
primers, nucleotides, and DNA polymerases used
in the PCR process) are removed from the solution
as it enters the pipette tip. Dispensing the solution
yields purified, high quality DNA ready to use in your
next application. You don’t need to use any capital
equipment, reagents or buffers, or enzymes — all you
need is a pipettor.

Diffinity RapidTip2 purifies PCR solutions and removes DNA polymerase
(standard, HotStart, and Proofreading) comparably to popular spin column
methods. PCR reactions were prepared and purified via a competitor’s
product (C), RapidTip2 (R), or left unpurified (U). Then these reactions were
placed in the thermocycler. Our sensitive assay generates a 600bp band
when polymerase is present and no band when polymerase is absent. The
unpurified (U) reaction contains a 600bp band that indicates polymerase is
present. The control and RapidTip2 lanes have no detectable 600bp band
indicating that polymerase was adequately removed.

• Optional: Microfuge tubes to store purified DNA.
Method
• Attach one or more of our pipette tips to a universal
or Rainin LTS® P100/P200 pipettor (single or multichannel) or compatible robot.
• Aspirate PCR product into the pipette tip and mix
by pipetting up and down for approximately one
minute.
• Dispense purified PCR product into a clean tube or
well and dispose of the tip in normal laboratory trash
— that’s it!
50bp-20bp-Diffinity RapidTip2 effectively removes dNTPs (not visible on gel), primers
(20bp), and primer-dimers (50bp). The PCR product was divided and subjected to PCR purification with either competitor’s products (C) or RapidTip2
(RT2: 1, 2, 3). RapidTip2 has high yield: the PCR amplicon (2Kb) shows no
loss of band intensity after purification.
