pBADFGH (D). The plasmid pBADFGH contained both operons of araBAD and araFGH, which
expression was controlled by the tac promoter individually. The repA and repB encoded the region
and proteins related to DNA replication in Rhodococcus strains. Other abbreviations of the plasmid
as shown: kan, kanamycin resistance; neo, neomycin resistance phenotype; ori, origin of
replication, tet, terminator and P, promoter.

Supplementary Fig. 2. RT-PCR amplification of araBAD mRNA from R. jostii RHA2 grown on
glucose or arabinose. M lane contains the 100-bp DNA ladder (NEB). The sequence of the primers
is listed in Table 2. The 143-bp PCR products amplified by using the primers araB1 and araB2
from RHA2 grown on glucose and arabinose were shown as lane 1 and lane 2. Similarly, lane 3
and lane 4 indicated the 117-bp PCR products produced by using the primers araA1 and araA2
from strain grown on glucose and arabinose. The 163-bp PCR products generated by using the
primers araD1 and araD2 were shown as lane 5 and lane 6. Lane 7 and lane 8 were the negative
controls of RT-PCR by using the template devoid of reverse transcriptase with primers of araB1
and araB2.
