Towards this goal we are in the process of doing the following: • Hired Anddre Valdivia in July to serve as research technician. He has already been trained to perform immunostaining, mouse transcardial perfusions, cryostat sectioning, plasmid extraction, and transformations. Anddre is in the process of training with Juli to gain expertise in working out protocols for optimization and testing different antibodies. His last training module will be focused on gaining expertise for performing and troubleshooting in situ hybridization. • The plan for the core tissue library is to have 4% PFA fixed tissues (using C57BL6 mice) at E13, E16, E19/P0, P4, P7, P14, P21, >P30 for immunostaining and in situ hybridization. A smaller collection of fresh frozen postnatal and adult tissue will be generated for select in situ hybridization projects (for greater sensitivity. • The MNAC can order a limited number of antibodies to test for your Conte project if it is likely that extensive mapping will be needed. Investigators can also send antibodies to the MNAC and the core will run pilot studies to optimize staining. • The Blakely Nlgn2 projects are serving as the core’s trial for in situ hybridization training. In September, Anddre will make new Nlgn2 probes. Anddre will do initial in situs with this probe. • Towards the end of year 01, Juli also will begin to introduce new in situ hybridization techniques in order to examine the distribution of splice variants and microRNAs. New methods are described in the literature, which Juli has begun to adapt.
