Cytokines and peptide growth factors including interferons and urocortin, for example,
transmit through intracellular signaling pathways which mediate transcriptional activity
and regulate cardioprotection. Transcriptional activity is regulated through a number
of intermediates in the cardiac myocyte and includes PKC and iPLA2/LPC pathways.
These two pathways can converge on the mitochondrion and help regulate
mitochondrial permeability either through the MPTP or the Kir6.1 channels (potassium
inward rectifier). Through phosphorylation activity, the AKT/mTor pathway regulates
protein synthesis (and subsequently cell volume - hypertrophy) and the MAPK kinase
pathway controls transcription of cardiac survival genes.

Cardiac stress such as I/R injury, ROS, or MI mediate transcriptional responses via the
STAT factors. STAT1 mediates cell death pathways, whereas STAT3 mediates
cytoprotection. Some inhibitors of the STAT pathways are highlighted: 1) U0126 and
PD98059 are pharmacological ERK inhibitors; 2) EGCG, the main constituent of green
tea can inhibit STAT1 tyrosine and serine phosphorylation; and 3) PIAS, SOCS and PTPs
are endogenous cellular inhibitors of the signaling routes critical in cardioprotection.

Cells were fixed with 4% formaldehyde in PEM buffer. The coverslip was incubated in blocking buffer of
5% powdered milk in TBS-T plus 0.02% sodium azide for 1 hour at room temperature. Blocking buffer
was removed and primary antibody was added at a dilution of 1/200 and incubated overnight at 4°C.
