Rat monoclonal [210] to Nicotinic Acetylcholine Receptor alpha 1 + 3 + 5
Specificity
This antibody was generated in rats against bovine and mouse muscle nicotinic acetylcholine
receptors (AChRs). It binds to the main immunogenic region (MIR) on the extracellular surface of
muscle AChR a1 subunit. This antibody binds to the alpha 3 and alpha 5 subunits of human
neuronal AChRs. The antibody binds the alpha 1 subunit weakly when the protein is denatured. It
can detect the alpha 3 and alpha 5 subunit native forms only, and therefore does not detect these
two subunits in immunoblotting.

Target
Relevance
The Nicotinic Acetylcholine Receptors (AChRs) are members of a superfamily of ligand gated
ion channels that mediate fast signal transmission at synapses. These receptors are thought to
be hetero pentamers composed of homologous subunits. The proposed structure for each
subunit is a conserved N terminal extracellular domain followed by three conserved
transmembrane domains, a variable cytoplasmic loop, a fourth conserved transmembrane
domain, and a short C terminal extracellular region. After binding acetylcholine, the Nicotinic
Acetylcholine Receptor alpha responds by an extensive change in conformation that affects all
subunits and leads to opening of an ion conducting channel across the plasma membrane.
