The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user. Application Abreviews Notes IP IHC-P IHC-Fr 1 Application Abreviews Notes ICC/IF Application notes ICC/IF: Use at an assay dependent dilution. IHC-P: Use at a concentration of 2 µg/ml. IHC Acetone-fixed Fr: Use at a concentration of 2 µg/ml. IP: Use at 2µg/mg of lysate. (Native and denatured) Staining of acid-alcohol fixed tissues requires antigen unmasking with 15,000U/ml of bovine testicular hyaluronidase in PBS, pH 7.4 for 30 min at 37°C. Not yet tested in other applications. Optimal dilutions/concentrations should be determined by the end user. Target Function Binding to cells via a high affinity receptor, laminin is thought to mediate the attachment, migration and organization of cells into tissues during embryonic development by interacting with other extracellular matrix components. Tissue specificity Found in the basement membranes (major component). Sequence similarities Contains 11 laminin EGF-like domains. Contains 1 laminin IV type A domain. Contains 1 laminin N-terminal domain. Domain The alpha-helical domains I and II are thought to interact with other laminin chains to form a coiled coil structure. Domains VI and IV are globular. Cellular localization Secreted > extracellular space > extracellular matrix > basement membrane.
