WB Use at an assay dependent concentration. Predicted molecular weight: 21 kDa. Use 0.5µg (or 0.5ng/lane). IP Use at an assay dependent concentration. ELISA Use at an assay dependent concentration. Use 0.5ng/well. 1 Application Abreviews IHC-P Notes Use at an assay dependent concentration. Perform heat mediated antigen retrieval before commencing with IHC staining protocol. Use 1µg. Target Relevance Human erythropoietin is member of the EPO/TPO family and encodes a secreted, glycosylated cytokine hormone composed of four alpha helical bundles. The protein is found in the plasma and regulates red cell production by promoting erythroid differentiation and initiating hemoglobin synthesis. This protein also has neuroprotective activity against a variety of potential brain injuries and antiapoptotic functions in several tissue types. It is produced by kidney or liver of adult mammals and by liver of fetal or neonatal mammals. Cellular localization Secreted Anti-EPO antibody images ICC/IF image of ab126876 stained HeLa cells. The cells were 4% formaldehyde fixed (10 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab126876, 5µg/ml) overnight at +4°C. The secondary antibody (green) was ab96899, DyLight® 488 goat anti-rabbit IgG (H+L) used at a 1/250 dilution Immunocytochemistry/ Immunofluorescence - for 1h. Alexa Fluor® 594 WGA was used to Anti-EPO antibody (ab126876) label plasma membranes (red) at a 1/200 dilution for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.
