Description and Main Properties 17.2 cm / 6.8" Pall PRC chromatography columns have been developed for fast selectivity and performance screening of chromatography sorbents. They guarantee optimal and consistent performance: l 1 mL and 5 mL prepacked columns, ready- and easy-to-use. The 5 mL columns are available in two different bed heights, refer to Table 1. l Direct connections to commonly used laboratory chromatography systems, e.g., ÄKTAu system (see Table 1). The 11.3 x 50 mm column is particularly suitable for connection to Pall’s Cadence™ BioSMB PD continuous multicolumn chromatography system l High packing efficiency l Fully scalable: once screened, the sorbent can be operated in the same conditions at either laboratory, pilot or production scale 2. It is composed of highly cross-linked cellulose beads and KANEKA’s proprietary alkaline resistant Protein A ligand. The cellulose base matrix has superior non-specific binding characteristics. KANEKA KanCapA sorbent allows high flow rates during affinity purification of IgG at development, pilot and commercial scale. Benefits: l Alkaline stability and long life cycle l Mild pH elution l High binding capacity l High flow rate operation and easy scale up from pilot to process An example of chromatographic conditions is detailed below. The optimal conditions depend on the characteristics of feed material. 1 mL (5 mm I.D. x 50 mm) 5 mL (8 mm I.D. x 100 mm and 11.3 mm I.D. x 50 mm) Highly cross-linked cellulose 65~85 μm Recombinant Protein A (alkaline resistant) Reductive amination ≥35 mg human polyclonal IgG/mL packed sorbent Stable in solvents commonly used in affinity chromatography pH 2~ pH 13 0.1~0.5 M sodium hydroxide, 6~8 M urea, 6 M guanidine hydrochloride Up to 500 cm/hr (bed height: 20~25 cm) ≥3 min (4~6 min is recommended) Slurry in 20% ethanol Slurry in 20% ethanol at 1~10 °C Table 2 Example of Chromatographic Conditions Flow rate Equilibration Load Wash Elution Strip Neutralization CIP Re-equilibration 100-500 cm/hr(1) (a residence time of 4 to 6 minutes is recommended) PBS, pH 7.4 (5 column volumes [CV]) IgG solution pH neutralized PBS, pH 7.4 (5 CV) Acetate buffer or citrate buffer, pH 3.5 1 M acetic acid (3 CV) PBS, pH 7.4 (3 CV) 0.1 M sodium hydroxide(2) (15 minute contact time) PBS, pH 7.4 (5 CV) (1) The flow rate corresponding to a residence time of 4 to 6 minutes depends on the column length. For columns of 5 cm height, 50-75 cm/hr flow rate gives 4-6 minutes of residence time. (2) 0.5 M sodium hydroxide can also be used (refer to Table 1) 1 mL column: 10x128 mm 5 mL column: 10x164 mm and 16x124 mm Body and end caps: Molded Polypropylene (PP) 17 μm frit: Polypropylene / Polyethylene (PP / PE) Built-in 10-32 fittings 1 mL column: 20 bar g (290 psi g); 5 mL column: 30 bar g (435 psi g) 3. Thermal Stability and Storage Temperature of use Storage temperature Storage solution between runs (1) 5% dynamic binding capacity is determined by frontal analysis at 3 minute residence time. (2) Alkaline conditions can be used for CIP but are unsuitable for long term exposure. (3) Prevent from freezing (4) For HPLC/MPLC/ÄKTA systems: direct connection. For systems equipped with 2 to 30 °C 2 to 8 °C 20% ethanol Column must never be frozen Column is shipped at ambient temperature ¼-28 or M6 connections: use a standard 10-32 male/¼-28 or M6 female adaptor (not supplied). 2 USD 3090a USD 3090a www.pall.com/biopharm 3
