Verification of Homogeneity during Liquid/Liquid
Mixing with the Pall® Jet Mixer
Mixing system: Pall Jet Mixer
Mixing biocontainer: 200 L Mixer biocontainer
Application mixing type: Liquid-liquid
The Pall Jet Mixer is one of the lightest and most compact single-use mixing systems. It
consists of a cubical shaped single-use biocontainer with a magnetically impelled turbine,
secured to the bottom of the biocontainer, working as a centrifugal pump.
The established 3D recirculation loop eliminates dead zones and ensures efficient and
fast mixing.
Introduction
Liquid-liquid homogenization is a common requirement in
biopharmaceutical processing, including after storage.
Furthermore, a cubical biocontainer shape is generally
considered to be preferable from a storage efficiency
(footprint) viewpoint. However, performing mixing in a cubical
biocontainer can be difficult; some mixing technologies can
be prone to uneven distribution within the mixing biocontainer,
or can even introduce unwanted particulate contamination.
The Jet Mixer impeller was developed to overcome these challenges.
In this experiment, a Jet Mixer was used to mix 200 L of salt solution. The objective was to
demonstrate how the Jet Mixer magnetic impeller is able to quickly and efficiently deliver a
high level of solution uniformity within the mixing biocontainer.
Experimental
A 200 L biocontainer was filled with 198 L of DI water. The Jet Mixer impeller was set to a
speed of 1000 rpm, and then 2 L of 3 M sodium chloride (NaCl) solution was added to the
mixing biocontainer in one of the top corners.
Solution homogeneity was monitored via conductivity readings taken at three locations
in the mixing biocontainer: (1) bottom corner opposite the injection point, (2) top corner
opposite the injection point, and (3) in the upper center of the mixing biocontainer
Results
Figure 1 shows the solution homogeneity in the biocontainer during mixing.
Mixing was 95% complete within 90 seconds, with steady state reached in 120 seconds.
