Unknown Bacteria by Jon Samel Or what the heck is that growing on Dr. Crawford’s laptop? • A swab was taken from Dr. Crawford’s laptop keyboard. • Growth on the plate showed two bacilli. • Successive attempts to separate the bacteria proved unsuccessful. Unknown Bacteria • Sample was pale white, round and convex. • Culture one was grown on Triptic Soy Agar at a temperature of 37 deg. Celsius. • Incubation time was 72 hours. Unknown Bacteria • Two bacteria located in the sample. One identified as Staphylobacillus by Dr. Crawford. • Initial simple stains with Methylene Blue did not stain plasma membrane but did stain the organelles. • Staining with Crystal Violet revealed single rods and clusters. Unknown Bacteria • • • • • Gram stain positive. Capsule stain negative. Acid fast positive. (Note results questionable) Catalase test positive. Endospore test shows sub terminal endospores. Unknown Bacteria • • • • • Starch test positive. Anaerobic test shows facultative anaerobic growth. Phenol red test shows pH but no gas formation. VP test A – Methyl Red positive. (Mixed acid) VP test B – Negative Unknown Bacteria • Mannitol Salt Agar shows --------• Four further attempts to isolate a pure culture proved unsuccessful. • Cultures were repeated in successively lower temperatures. *37, 34, 32 and 20 deg.
