Which single step is the most important?
If you use the wrong stain or it doesn't quite work you can section and stain more tissue.

The rough and the smooth endoplasmic reticulum, Golgi apparatus, Cell membrane, lysosomes.
Cytoplasm?
Made of main water. It holds together the structure of the cells in place and protects them from being damaged.
