Building the Backbone In the nucleus you have nucleotide triphosphates dTTP, dATP, dGTP and dCTP If you break off 2 phosphates get a huge amount of Energy released (just like when the cell uses ATP in other reactions) To build the backbone DNA Polymerase takes a nucleotide triphosphate minus 2 of the phosphates and uses the energy to build the phosphodiester bond between the OH on the 3’ carbon of the nucleotide above to the P on the 5’ carbon from the new nucleotide (the one that has lost the 2 phosphates).
