The scientific word journal(communicated) Summary of thesis This research work was conducted in two parts to tackle the following problem of scalp/ hair: 1. Yeast like fungi Malassezia which causes dandruff in human beings. 2. An insect, human head louse, which is an ectoparasite and causes itching of scalp. The present study discussed techniques and media used to isolate, maintain, preserve, and identify the 13 Malassezia species. Broth media was developed as a result, for antifungal susceptibility testing as well as culturing of the test fungus i.e. Malassezia. These findings are now under consideration for Patent. Our results bring new information to the literature data about the antidandruff activity of plant extracts and Curcuma longa and Coriandrum sativum essential oil against the Malassezia. In-vitro activity of market available synthetic (Ketoconazole Fluconazole, Clotrimazole, Sertaconazole) was also determined against Malassezia spp.In silico studies in form of phylogenetic analysis of four common pathogenic Malassezia spp. which has been tested throughout the study was done using CLASTALW (EBN).Extraction of the DNA of Malassezia spp. was done by Bioneer Genomic DNA Isolation Kit and quantification of DNA was also performed by RT-PCR (Rotor gene 6000). In present study, Pediculocidal activity of botanicals was determined by contact toxicity as well as fumigant bioassay. All these results were statistically analyzed by ANOVA, χ2 Test using Minitab software as well as manually. Antioxidant potential of selected botanicals was also examined for value addition to the formulation developed. On the basis of their ingredients, two formulations have been developed.
