DOB 22/07/76. My research background consists mainly of research in the field
of genetics: Sequencing, SNP scoring, linkage analysis, expression and protein
studies. Diseases studied: COPD, asthma, T1 Diabetes and Multiple Sclerosis.
I have become increasingly interested in studying the environmental factors
underlying poor health, particularly from a dietary standpoint.

Project description
Bacillus anthracis, a gram-positive, nonmotile, spore-forming, rod-shaped
bacterium, is the etiological agent of anthrax.

Fully virulent strains of B. anthracis carry two large plasmids, pXO1 and

pXO2, encoding the virulence factors respectively toxins and the capsule.
The complete DNA sequences of pXO1 and pXO2 as wall as the cellular
model of action of anthrax toxins are known (Mock M, Fouet A. Anthrax.
Annu Rev Microbiol 2001;55:647-71).
The studyes of atypical strains of Pasteur vaccine (n. 1, n. 2-H and n. 217JB) and of the italian attenuated B. anthracis strain, named Carbosap,
revealed the presence of both plasmids pXO1 and pXO2 (Fasanella A,
Losito S, Trotta T, Adone R, Massa S, Ciuchini F, Chiocco D. Detection
of anthrax vaccine virulence factors by polymerase chain reaction.
Vaccine 2001 Jul 20;19(30):4214-8).
The use of Suppressive Subtractive Hybridization (SSH) between virulent
and typical vaccine strains (pXO1-/pXO2+ or pXO1+/pXO2-) or atypical
vaccine strains (pXO1+/pXO2+) and the screening of subtracted cDNA
libraries, could be useful in the isolation of the genomic differences
between virulent and vaccine strains and in the comprehension of
protectiveness mechanisms of vaccination.

Title of fellowship project Neuromodulation of vagal nerve activity for the treatment of obesity
to be carried out abroad
Keywords
Obesity, vagal nerve, neuromodulation, electrical stimulation, GI motility
Specify the field of main
Experimental and clinical study on GI motility and food ingestion control
research activity and results Please mark the keywords on the attached list that relate to your activities
Objectives:
Project description
- To develop minimal invasive, accurate, safe and cost-effective
Including -if possible innovative aspects and
method of electrode implantation into gastric wall.
expected results
- To develop evidence based parameters for adequate vagal
response.
- To evaluate effect of permanent neuromodulation of the vagal
activity on body mass.
- To search for pathomechanism.
- To evaluate safety of the method and incidence of adverse effects.
What's new:
Percutaneous electrode placement by endoscopic assistance. - (less
invasive)
Evidence based stimulation parameters - (increased efficiency)
Stimulation depended on food intake - Increased stimulation
response to both food and liquids intake (no restrictions to high
caloric fluids).

No risk of anastomotic leaks and low for other perioperative
complications.
Anatomical passage is maintained - no risk of nutritional
deficiencies (vitamins, iron etc) and cholelithiasis.
General description:
A. Experimental studies - 12 months
2 months - development of endoscopic method of electrode
placement. (series of phantom and animal experiments)
4 months - development of stimulating current parameters.
6 months - long-term study on influence of permanent vagal
neuromodulation on body mass.
B. Clinical studies - 12 months
Development of equipment and preparation for pre-clinical study in
human patient.
In case of positive, promising and sufficient experimental testes of
1st phase- initiation of clinical study.
Evaluated parameters:
Food intake - daily,
General condition - twice a week,
Body mass - weekly,
Morphometry - weekly,
Parameters control - weekly,
ECG with HRV analysis - monthly,
Blood tests - blood cell count, GOT, GTP, albumines - monthly,
Hormones - systemic blood - CCK, leptins (fasting and mealtime) monthly.
Metabolism - BUN, creatinine, uric acid - monthly
Upper GI endoscopy - at establishment and at completion of the
study (ulcer formation, electrode position)
