Product Specification gRAD-RWB For use with Rabbit antibody-derived detection conjugates to analyse Whole Blood samples Summary In most applications, the specific test reagent contains both a biotinylated capture reagent and a detection conjugate. When the test reagent is mixed with the sample, both the capture reagent and detection conjugate react with the analyte present in the sample to form a complex. The sample mixture is applied to the round well of the gRAD, blood cells or other particles are filtered out and the liquid flows through the device, passing the test line. The test line contains a biotin-binding protein that captures both complexed and free biotinylated capture reagent, the ratio of the first to the second relating to the amount of analyte present in the sample. Sufficient capture reagent is present in the test reagent to saturate the test line after running for 5-10 minutes. On saturation, the amount of analyte complex immobilized at the test line depends on its ratio to the free capture reagent, not on the volume of sample that has crossed the test line. The round and square application wells Apart from the round well, the gRAD has an additional square well which can be used when the detection conjugate is inhibited by other components present in the sample (e.g. immune response assays in which the detection conjugate binds all antibodies or a class of antibodies). In these cases the sample is mixed with the biotinylated capture reagent before being applied to the square well, whereupon the detection conjugate is added to the round well. The detection conjugate solution pushes the sample mixture in front of it through the gRAD to the test line, where the biotin binding protein captures the biotinylated capture reagent. The rest of the sample mixture containing the inhibiting components continues through the gRAD, leaving the capture reagent-analyte complex at the test line ready to react with the detection conjugate that passes over it. Control line - for rabbit antibody-derived detection conjugates The gRAD-RWB is designed for use with rabbit antibody-derived detection conjugates in that the control line contains antibodies that bind the mouse antibody which is responsible for immobilizing the detection conjugate at the test line. A visibly coloured control line indicates that the sample mixture or sample mixture + detection conjugate has passed correctly through the device. Filter for whole blood or particulate samples The gRAD-RWB is designed with a filter to permit the analysis of whole blood and samples containing particulate matter. The gold colloid conjugate passes through the filter with the rest of the sample mixture to interact with the test and control lines. Supply The gRAD-RWB is supplied in a plain white box of 40 units marked only with the batch number. All other labelling is present on a plastic film wrapping that is easily removed, allowing you to put on your own labels as required. Each unit is packed in a white foil pouch which is also marked only with a batch number to permit your own labelling. The batch number is printed directly on the gRAD, as well as an broad arrow indicating the direction in which the gRAD should be placed in reader.
