User Guide for HP 8453E UV-visible Spectroscopy System
All USERS MUST SIGN IN THE USER BOOK BEFORE USE!
Steps:
1. Turning on the HP 8453E UV-visible Spectroscopy by pressing the line power
switch. The indicator at the upper right corner of the front panel will light with
three colors. The yellow color means start up, the green color means ready to
measure, and the red color means an error condition.
2. The control panel screen will show Tasks (Start here).
3. Press F1-key (Tasks (Start here)) on the screen.
4. The screen will show the following options,
(1) Single WL
(2) Multi WL
(3) Equation
(4) Spectrum
(5) Quantification
(6) Kinetics
5. Use arrow keys in the control panel to select the option you need.
6. For simple UV spectrum measurement, please select Spectrum, and press Enter.
7. The screen will show the Data type you need, use arrow keys to select the data
type. For most application, you can select Absorbance.
8. Press the F6-key (Done) to save your settings and exit the sreen.
9. Put your solvent in the Quartz Cell, wipe off all fingerprints and solvent out of the
cell, and insert the cell into cell holder. Lock the sample cell in place by pushing
the locking lever point downward. The clear sides of Quartz Cell should face the
UV light source. Use only Kimwipes or lens tissues to wipe the surface of the
Quartz Cell, and never dry the inside of a Quartz Cell with lens tissues.
10. Press the F8-key (Blank) to make a measurement on your solvent.
11. Remove the Quartz Cell from cell holder, clean the cell and put your sample into
cell.
12. Press F7-key (Sample) to make a measurement on your sample.
13. Remove the Quartz Cell from cell holder, clean the cell and put the cell upside
down on a piece of Kimwipes for drying.
14. If you need to read the absorbance value of a particular wavelength, press F6-key
(Graphic), the whole spectrum will zoom in, use arrow keys to move the cursor in
the spectrum to the wavelength you need, the reading is shown directly in the
screen.
15. Never turn on and off in UV spectrometer more than two times in a day.
