Abstract No: 50 EFFECT OF KIDNEY HERBS ON SECRETION REGULATION OF PRL AND TH CELL CYTOKINE IN DECIDUE OF EARLY PREGNANCY FROM BROMOCRIPITINE-INDUCED ABORTION 1 1 G. Suiqi, 1L. Kunming, 1J. Lihui, 2L. Yiwei Department of OBS/GNY, 2Physiology and 3Immunology, Hospital of Fudan University, China Objective: To study regulation mechanism on the field of endocrinology and immunology with Kidney-replenishing herbs for spontaneous abortion. Methods: The model to induce early abortion by taking Bromocriptine (0.125 mg/d,sc) during pregnant day 6-8 was reformed. 5 groups were divided: model rats + Herbs ig (group A); model rats + distillate water ig ( group B) ; model rats + PRL sc (group C) ; normal pregnant rats +75% alcohol sc ( group D); normal pregnant rats + distillate water sc ( group E); non-pregnant rats (group F ).Both rats of 5 groups at pregnant day 12 and non-pregnant rats on estrous cycle were killed ,and pregnant outcome were evaluated. Levels of serum PRL, P on the day 4,7, 10, 12 of pregnant rats and estrous cycle of non-pregnant rats were measured by RIA ,Expression levels of both PRLR,PR, of decidue / ovary / endometrium and Th type-l (IFN-?,TNF-? ) / Th type-2 (IL-4, IL-10 ) of decidue were detected by RT-PCR . Results: Abortion rates in group A, B, C, D, E were 22.22%,70.83% (group A vs B ,P<0.05), 0%,8%,4% respectively. Pregnancy rates in group A, B, C, D, E were 77.78%,29.13% (group A vs B ,P<0.05),100%,92%,96% respectively. The number of embryo in group A,B, C,D,E were 234, 64 (group A vs B, P<0.001),153,242 , 262 respectively. Levels of serial serum PRL, P of group A,B,C,D,E of during pregnant day 4,7,10,12 were higher than that of non-pregnancy rats(group F) at estrous cycle (P <0.001); Levels of serum PRL,P of group A,C,D,E during days 7,10,12 were higher than that of group B during same days (P <0.00I); Expression levels of PRLR, PRL mRNA, PR, P mRNA in decidue / ovary of group B were lower than that of groups A,C,D,E (P<0,05), but there are no significant differences between group B and group F(P>0.05). The transcription levels of IL-4 mRNA,IL-10 mRNA in decidue of group A were higher than that of group B [ (64.81±3.07 vs 22.21±2.42; 46.56±3.83) (ng/100?l ) (P< 0.05) ]; .The transcription levels of TNF-?mRNA, IFN-? mRNA in decidue of group A were lower than that of group B[ (27.52±6.16 vs 51.19±3.56;17.20±1.13 vs 21.54±2.16) (ng/100?l) ( P< 0.05)]; Ratio of IL-10 / IFN-? in decidue of group A were higher than that of group B (2.72±0.29,1.05±0.13) (P< 0.05) which showed deviation Th2 type cytokines in balance of Th2/Th1 between group A and B. The ratio of Th2/Th1 with P level comparing to PRL was positive correlation by partial correlation coefficient(Pearson) (?value 0.64, P<0.05). Conclusion: Rat model is characteristic of dysfunctional regulation on endocrine-immune from Bromocriptine-induced early abortion. Serum PRL secretion levels were decreased and inhibited the expression levels of PRLR / PR mNRA in both decidue and ovary in the Bromocriptine-treated rats and secondarily trigger a predominate Th1 type cytokine, resulting in embryonic resorption in pregnant rats. Kidney Replenishing Herbs could regulation function of endocrine- immune to reserve the effects by restoring serum PRL,P levels or by increasing expression of PRLR / PRmRNA in decidue and ovary,as well as the bias response for Th2 type cytokine of decidue resulting in success pregnancy.
