No extraction DNA extraction – specifically useful for genotyping
Lysis-Buffer:
1x PCR-Buffer (0.5mM MgCl2)
0.045% Tween-20
0.045% NP-40
50µg/ml Proteinase K
Earpunches/Small parts of tissue are incubated at least 4h in Lysis buffer
(100µl/piece) at 56-65°C, then the Proteinase is inactivated by heating to 95°C for at
least 10min. short spin. Use 1-2µl of lysate for PCR. Long-term storage of samples
requires freezing.
It's easiest if you make lysis buffer aliquots of 990µl and keep them frozen at -20. add
10µl of proteinase K(50µg total), just before using and then split the 1ml between 10
tubes.
(tissue can stay in lysis buffer also o/n).
