Instituto de Fisiología Celular, UNAM. lilianaq@cinvestav.mx Alzheimer's disease (AD) is the most common cause of dementia in the world and affects more than one million elders in the world. One of the hallmarks of AD is the formation of amyloid plaques composed of the protein fragment (39 to 42 amino acids) beta-amyloid (BA). BA is known to coordinate heavy metals (Cu, Fe, Zn) with high affinity and is thought to be involved in the generation of reactive oxygen species. This study is focused on the characterization of different BA-Cu(II) complexes using spectroscopic techniques like electron paramagnetic resonance, UV-Vis absorption and circular dichroism under different conditions (pH, Cu concentration, etc). In the fragment BA(1-16), we have found two sites of coordination to Cu, which are dependent on pH. We have evaluated the participation of the amino terminal group in Cu coordination studying the acetylated form of the BA peptide. We are also evaluating the redox chemistry of BA-Cu complexes using electrochemical techniques like cyclic voltammetry (CV).
