Aim
The aims of the experiment were to observe how a light microscope illuminates the image of
a specimen, to find the field of vision under a 40x objective and then to estimate the size of
cells that are examined and also to determine whether the bacteria are gram positive or
negative.
Methods
Determination of the FOV and Observation of The Buccal Smear
To determine the field of view at 40x magnification, a buccal swab was taken from the inside
of the cheek, located onto a hemocytometer and a drop of methylene blue was added into it.
This sample was covered with a cover slide and placed onto the microscope for observation.
The diameter of the viewed circle was measured by first counting how many edges of the
square were fitted into the diameter, 9 counted, and then multiplying this number with the
known length of one edge of the square which was 0.05 mm. Therefore, the diameter was
found as 0.45 mm (9x0.05mm) then the radius was 0.225 mm. After that the epithelial cells
were found by adjusting the stage and observed. They were fitted into 1.5 square.
