The purpose of this lab is for you, the student, to learn how to view a prepared slide using a compound light microscope.! ! Materials! ! 1 compound light microscope! ! 2 prepared slides! ! Procedure! 1. With your partner, collect one microscope, and one prepared glass slide. Always carry the microscope with one hand holding the arm and one hand underneath the base!! 2. Set the microscope up carefully on a counter close to an outlet. Remember to keep the microscope away from the edge!! 3. Plug the microscope in. Check that the stage is completely lowered and that the low power objective lens is in place.! 4. Place your slide in the slide clips. Turn on the light source. ! ! ! ! 5. Look through the ocular lens (eye piece) at the specimen. Use the coarse focus to bring the stage up until the specimen is focused.! 6. Once you have used the coarse focus to view the specimen, use the fine focus to make the image as clear as it can be. At this point, the fine focus knob is the only knob that needs to be touched.! 7. Try turning the diaphragm to see what the specimen looks like with different amounts of light shining through it. ! ! 8. You and your partner should both draw what you see in your field of view. Now, turn the nosepiece so the high power lens is in place. Using the fine focus knob only, adjust the focus so that the image is clear. Draw what you see in your field of view. Return your first slide to the slide case and choose a second specimen to look at. Make sure you reset your microscope so the stage is lowered all the way down and the low power objective lens is in place.! 11. Repeat steps 4-9 with the second specimen. When putting your microscope away, return the objective lens to the lowest objective, completely lower the stage, carefully wrap up the cord, cover the microscope, and put it safely away as directed.
