Salah satu cara untuk mendapatkan enzim amilase dari bakteri yaitu dengan melakukan isolasi bakteri dari produk hasil fermentasi amilum. Salah satu sumber makanan yang menghasilkan karbohidrat adalah umbi gadung. Gadung mengandung karbohidrat dengan kadar sebesar 18%, lemak 0.16%, protein 1.18%, serat kasar 0.93%, dan kadar abu 0.69%. Berdasarkan hasil isolasi bakteri yang berasal dari umbi gadung paska fermentasi 72 jam didapatkan 4 isolat bakteri murni yaitu : FDH-1, FDH-2, FDH-3, dan FDH-4. Setelah dilakukan isolasi dan identifikasi koloni serta dilakukan uji aktivitas hidrolitik menggunakan media Starch agar isolat FDH-1 dipilih untuk isolasi dan identifikasi molekuler untuk dilanjutkan isolasi DNA, PCR, elektroforesis yang kemudian diproses BLAST dan pensejajaran hubungan kekerabatan. Isolat FDH-1 memiliki cabang yang sama dengan Pseudomonas stutzeri strain Os. menunjukkan isolat FDH-1 memiliki hubungan kekerabatan yang sama Pseudomonas stutzeri strain Os. Berdasarkan hasil yang diperoleh melalui produk fermentasi umbi gadung dengan demikian strain bakteri Pseudomonas stutzeri diberi nama isolat bakteri Pseudomonas stutzeri strain FDH-1. One way to get the amylase enzyme from bacteria is by isolating bacteria from the fermented starch products. One source of food that produces carbohydrates is the gadung tuber. Gadung contains carbohydrates with content of 18%, 0.16% fat, 1.18 % protein, 0.93% crude fiber, and 0.69% ash content. Based on the results of bacterial isolation from gadung tubers after 72 hours of fermentation, 4 pure bacterial isolates were obtained, namely: FDH-1, FDH-2, FDH-3, and FDH-4. After isolation and colony identification were carried out and hydrolytic activity tests were carried out usingmedia Starchso that the FDH-1 isolate was selected for isolation and molecular identification to be continued with DNA isolation, PCR, electrophoresis which was then processed by BLAST and alignment of kinship relationships. FDH-1 isolate has the same branch as Pseudomonas stutzeri strain Os. showed that FDH-1 isolates had the same kinship relationship as Pseudomonas stutzeri strain Os. Based on result obtained through a product fermentation of gadung, a strain of bacteria Pseudomonas stutzeri is named a Pseudomonas stutzeri strain FDH-1.
