Converting from linear flow (cm/hour) to volumetric flow rates (ml/min) and vice versa
It is convenient when comparing results for columns of different sizes to express flow as linear
flow (cm/hour). However, flow is usually measured in volumetric flow rate (ml/min). To convert
between linear and volumetric flow rate use one of the formulae below.
From linear flow rate (cm/hour) to volumetric flow rate (ml/min)
Volumetric flow rate (ml/min) =
=
LFR p x d
x
60
4
Linear flow rate (cm/h)
2
x column cross sectional area (cm )
60
2
where
LFR = linear flow rate in cm/h
d = inner diameter of the column in cm
From volumetric flow rate (ml/min) to linear flow rate (cm/hour)
Linear flow rate (cm/h) =
=
Volumetic flow rate (ml/min) x 60
2
column cross sectional area (cm )
VFR x 60 x 4
2
pxd
where
VFR = volumetric flow rate in cm/h
d = inner diameter of the column in cm

Recommended flow rates for the different cartridges:
cartridge volume
1 ml cartridge (2-1233-001)
5 ml cartridge (2-1234-001)
physical dimension
5 mm (i.d.) x 50 mm
11.3 mm (i.d.) x 50 mm
flow rate
1 ml/min or 0.3–1 drops/second*
3 ml/min or 1–3 drops/second*
*The drop/sec value depends on several parameters (diameter of the outlet, viscosity of the solution etc)
and, therefore, has to be determined one-time for each individual setting. Since column bodies are highly
pressure resistant (“H-PR”) and can tolerate 20 bar the columns can be used with a flow restrictor.
Important notes:
Strep-Tactin® resin is located in the lower part of the cartridge body only.
Cartridges do not generate significant back pressure which makes the use of flow restrictors superfluous. Therefore,
IBA recommends not to use flow restrictors to avoid inhomogeneities resulting from buffer changes during
chromatography.
