Having resided in different countries gave me exposure to the various facets related to the environment, where various regions had differing attitudes and policies towards their local environment. From the abundant tropical rainforests of Borneo islands to the arid regions of the Middle East and teeming cities of India, energy requirements are varying and so are their impacts on the environment by harnessing them. Coupled with my interest in sciences and technology, it inspired me to strive for a career in the development of green technologies. My career goal is to see the implementation of these technologies for achieving sustainability in the society. By the utilization of nature’s energy resources, achieving minimal or no impact on the environment must be the goal for all the industries in the coming decades. Project mentors: Dr. Annemiek ter Heijne & Mr. Fei Liu (PhD candidate, on-site supervisor) Bachelor Thesis, Chemical Biology laboratory, Centre for Biotechnology, Anna University December 2011-April 2012 Aim of project: Isolation and Screening of Microorganisms from Sludge used in a Microbial Fuel Cell. Work done MFCs were constructed to check for the voltage generated. Biofilm formation was observed on the electrodes. Screening of the biofilm-forming microorganisms was done. Based on their morphology, two types of colonies (white and red) were identified. When these cultures were inoculated in separate MFCs comprising autoclaved sludge it was found that they produce a higher voltage of 628 mV and 860 mV respectively which is higher than the voltage produced by the control by 71 mV and 303 mV. A separate experiment was devised to amplify the exoelectrogenesis of the mixed culture of microorganisms in the anaerobic sludge using the Urey-Miller Setup. After a run time of 14 days in the setup, the moisture sample deposited on the walls of the reactor was cultured. One colony was identified. This culture was then inoculated into separate MFCs along with the modified sludge from the reactor and the voltage output was measured. The voltage output in the modified sludge was found to be 931 mV, 1.7 times greater than the unmodified sludge (control) indicating the purpose. Work done We synthesized porphyrin by Rothemund synthesis and Lindsey’s synthesis. It was sulphonated using chlorosulphonic acid and dichloromethane. The zinc complex of the sulphonated porphyrin was synthesized. The Zn-complexed sulphonated porphyrin was incorporated in the bacterial cell membrane (Escherichia coli used) and was used in fluorescence studies. Result: Fluorescent activity of the Escherichia coli was observed under the fluorescent microscope and enumeration of microbial consortia was done.
