Evonik’s HPAPI Custom Manufacturing
Advantage for Highly Potent Oncology Drugs
Compounds to treat cancer are typically known as highly potent active pharmaceutical ingredients
(HPAPIs) because they generally act at very low concentrations. High potency places substantial
additional requirements on the handling and formulating of HPAPIs. In addition to FDA regulations,
the manufacturer must comply with a large number of worker safety and environmental regulations to
ensure the protection of employees and the environment across areas such as the laboratory, production,
warehousing, and waste treatment facilities. This requires an expertise in regulations, industrial hygiene,
process development, equipment design, as well as materials handling and waste treatment.
Evonik operates total reactor volumes of 135 m3 for ton quantities and 35 m3 for kilogram or
development quantities of HPAPIs. Containment technologies can meet Occupational Exposure Levels
of < 0.1 μg/m3 air (8 hour TWA). In addition, Evonik can go from laboratory to full scale manufacturing

CML Invests to Support Late-Phase and Commercial API Demand

Many contract API suppliers who have a strong base
of assets to handle programs from phase 2 through
launch have seen remarkable growth in their portfolio
of API projects. And Cambridge Major Labs is no
exception. In July of 2012, CML embarked on a major
expansion of its large scale API plant in Wisconsin.
Scheduled to be complete in April, the expansion will
increase CML’s large scale capacity by nearly 30 percent,
enabling more capacity for the large number of phase 2
and phase 3 APIs in CML’s funnel. Having a supplier that
can meet the growing volume needs as APIs progress
to commercialization is key to a successful new product
launch; no technical transfer is required, and the API
supply is effectively de-risked.
