VersaFlashTM Solid Sample Cartridge Kits

c) Once the desired volume of adsorbed media is in the solid
sample cartridge, backfill with non-adsorbed packing media to
within 0.5 cm from the top of the cartridge. Attach frit insertion
sleeve and use frit insertion plunger to level the packing media,
do not over pack the media.

The solid sample cartridge kits are designed for samples that have
low solubility. Both liquid and solid samples with low solubility can be
dissolved in a small amount of strong solvent and absorbed onto an
inert media, such as silica or celite. The solvent is evaporated and the
absorbed media is packed into the solid sample cartridge. The
samples thus can be desorbed onto the separation cartridge in the
VersaFlash station. The materials of construction of the solid sample
module are polypropylene, stainless steel, and Teflon®.
The solid sample cartridges should be used at operating pressures
less than 50 psi.

d) Once the packing is leveled, place frit (smooth side up) into the
frit insertion sleeve. Use the frit insertion plunger to press the frit
into place, this will set the proper depth for the frit.

Loading a Solid Sample Cartridge
If the sample material exhibits low solubility in the mobile phase to be
used in the flash purification, it may be necessary to apply the
sample through the use of a solid sample module using the following
procedure:

a) Adsorb the sample material onto a solid support media such as
silica or celite. The media is then dried along with the adsorbed
sample material.
b) Pour the desired volume of adsorbed media into the appropriate
sized solid sample cartridge.

e) Remove the frit insertion sleeve and inspect the cartridge. Wipe
any excess packing from the top edge of the cartridge.

f) Attach the stainless steel cap and hand-tighten. Approximately
4 mm of thread will be visible on the tube; this is normal. The solid
sample cartridge is ready to desorb by placing a VersaPak™
cartridge after the solid sample cartridge using the cartridge
stacking connector. Follow instructions for cartridge stacking in
the VersaFlash station instruction manual.
g) The stainless steel cap should be removed and cleaned directly
after use. Under prolonged conditions, some organic solvents
could swell the cartridge causing difficulty removing the stainless
steel cap. If this occurs, flush the solvent from the cartridge and
let it soak overnight in a non-aggressive solvent such as water
or methanol to reduce the swelling. The stainless steel cap is
machined for use with a 13/16" wrench if the cap cannot be
removed by hand. Never use a wrench to tighten the cap onto the
cartridge.
h) It is recommended that you periodically inspect and replace the
O-ring inside the stainless steel cap. To remove the worn O-ring,
simply use forceps or a small spatula to pry the O-ring from the
groove. Care should be taken to not damage the Teflon flow
distributor or groove in the cap. To replace the O-ring, lay the new
O-ring inside the cap. The O-ring should be pressed into the Oring groove prior to screwing the cartridge on. Use a dull object
to press it into place and to minimize the potential for damage to
the O-ring. Screw the empty solid sample module into the cap to
seat the new O-ring. Additional replacement O-rings are available. (See Ordering Information).
i) The solid sample cartridge tubes are intended for one time use,
they are not recommended for reuse, due to concerns of sample
cross contamination.
