Abcam’s anti-SS-B IgG Human in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is
designed for the accurate quantitative measurement of IgG class antibodies against SS-B in
Human serum and plasma.
A 96-well plate has been precoated with SS-B antigens to bind cognate antibodies. Controls or
test samples are added to the wells and incubated. Following washing, a horseradish
peroxidase (HRP) labelled anti-Human IgG conjugate is added to the wells, which binds to the
immobilized SS-B-specific antibodies. TMB is then catalyzed by the HRP to produce a blue
color product that changes to yellow after adding an acidic stop solution. The density of yellow
coloration is directly proportional to the amount of SS-B IgG sample captured in plate.

Relevance
SS-B/La, an ubiquitous nuclear protein of 46-48 kD, is a target antigen of autoantibodies in SLE
and Sjogren's syndrome and is involved in the maturation of RNA polymerase III transcripts. SSB/La shuttles between nucleus and cytoplasm and exists both free and as a component of the
SSA/SSB ribonucleoprotein cytoplasmic particle. Autoantibodies to SSB are detected by
ELISA in 25% of SLE, in 80% of subacute cutaneous lupus, in the majority of infants with
complete heart block and in 70-90% of primary and 50% of secondary Sjögren’s syndrome.
Sjögren's syndrome is a chronic inflammatory autoimmune desease, characterized by
destruction of exocrine glands (minor salivary glands, lacrimal glands, parotid) mediated by T
lymphocytes. It has a frequency of 1%, mainly affects women (9:1) over the age of 50 years, and
seems to be associated with a genetic predisposition related to HLA-DRw52 (which increases
the risk of developing disease by a factor of 20) and HLA DR3 genes and probably to viral
infection by EBV and HTLV-1. SS also produces an excessive activation of B cells with
production of autoantibodies such as rheumatoid factor (RF), anti-SS-B/Ro, anti-SS-B/La. In
patients with SS these antibodies present in 60% of cases when tested by immunodiffusion and
up to 90% when tested by ELISA, and they are associated with more frequent extraglandular
involvement. They may also be present in approximately 30% of patients with SLE. High titers in
ELISA, especially for anti-Ro 52, are a risk factor for neonatal lupus.
