CLAUSEN MEDIUM Product Number C0596 Product Description Clausen Medium is recommended for sterility testing. This medium is a very nutritious medium consisting of casein enzymic hydrolysate, papaic digest of soyabean meal, yeast extract and dextrose. L-cystine and thioglycollate act as reducing agents. The metals help isolate anaerobic sporeformers. Tween 80 is added to this medium to overcome the effects of cationic agents (in combination with lecithin already present in the formulation) which can exert a bacteriostatic effect in vitro. This medium is yellow colored and clear in appearance. Under aerobic conditions it turns pink. Therefore at the time of use the upper third of the medium should be pink. Preparation Instructions Suspend 40 grams of Clausen Medium in 1000 mls of distilled water containing 3 grams of (P 8074) Tween 80 and 5 grams of (G 6279) Glycerol. Heat to boiling to dissolve the medium completely. Dispense as desired and sterilize by autoclaving at 15 lbs. pressure (121 °C) for 15 minutes. Place in a cool, dark place until use. Do not resterilize the medium. Storage Store the dehydrated medium at 24 °C. The prepared medium should be stored in a cool place (NOT 4 °C) and in the dark for not longer than one month. Product Profile Appearance Yellow colored, homogeneous, free flowing powder. Color and Clarity Light straw colored, clear to slightly opalescent solution with the upper 10% of the solution pink upon standing. Cultural Response Cultural characteristics are observed after 18-48 hours at 35 – 37 °C.
