Thank you for your order of TargetSeq™ Custom Enrichment Kits. Please complete this form to ensure accurate
processing of your order. If you have questions, contact targetseqcustomorders@lifetech.com.
Background information
• TargetSeq™ Custom Enrichment Kits provide a convenient, high-quality method to enrich for targeted
genomic regions.
• The researcher specifies the species, genome build, and target region(s). Based on the regions selected by
the researcher, Life Technologies will design capture probes using proprietary algorithms, and send the
design proposal to the researcher for approval. After the design has been approved, Life Technologies will
produce the custom capture probes and ship the kit to the researcher.
• When selecting region(s) of interest, please keep in mind that TargetSeq™ Custom Enrichment Kits can
capture 100kb to 60Mb genomic regions. These regions can be contiguous, such as disease or trait
associated regions, or noncontiguous, such as exons of a candidate gene panel. Life Technologies currently
chooses probes only against unique parts of the genome, as defined by proprietary repeat-masking methods.
Thus, repetitive portions of the region in which you may be interested might not be covered by the design.
You will receive design files, showing regions covered by the capture probes, for approval before the final
product is manufactured.
• If you need multiple designs, please use a separate form for each design.
• The performance of targeted enrichment methods and products may be unpredictable due to population
variability among genomes and other properties intrinsic to the targeted region(s). Examples of such
factors include: target sequence complexity (e.g. G/C content), repetitive element distribution (e.g.
retrotransposons), evolutionary history of the targeted region (e.g. conserved pseudogenes and gene family
relationships), and population variation from the reference genome (e.g. CNV and hypervariable regions).
When using a previously untested capture design, we recommended performing a small-scale experiment to
determine the capture characteristics of a specific target, before beginning a larger study.

Human genome build:
Specify the human genome build from the UCSC Genome Database
(http://genome.ucsc.edu/) to which your coordinates apply.
Design description:
Specify a brief description of your design (e.g. “Sequence capture design for cancer genes”).
Capture target size:
Using the information in the coordinates file that you created, specify the total size of your
capture target (in bp).
4. Generate a coordinates file
A coordinates file is required to process your TargetSeq™ Custom Enrichment Kit order. This file should
contain the set of coordinates for your regions of interest. The coordinates file is a tab-delimited text file (.txt)
that stores coordinate data in a three-column format, where each row of data contains the chromosome
(column 1), start position (column 2), and stop position (column 3). Prior to submitting your file, verify that
your coordinates match the genome build in the UCSC Genome database (http://genome.ucsc.edu/). For more
information on generating a coordinates file, see the TargetSeq™ Custom Enrichment Kit Ordering Guide (Part
no. 4471870).
IMPORTANT! You must supply the exact coordinates of each region; functional descriptions of your target
regions are insufficient.
Note: For small target regions of <100bp in size, Life Technologies extends the regions to 100bp to increase
capture efficiency.

5. Provide additional information
Include any additional information about your order that could help the designer.
6. Submit your order
After you complete this form and have generated your coordinates file, submit the form:

a. After you have completed all fields, click Submit
Form to save this form and attach it to an email.
Note: The email created by this form will address itself to Life Technologies.
b. Attach the following files to the open email:
• Coordinates file (see step 4)
• Purchase order file (a scan or a PDF of your purchase order)
c. Send the email containing your completed order to Life Technologies.
Note: If you do not have immediate access to an email application, then save and send this completed form,
your coordinates file, and your purchase order to targetseqcustomorders@lifetech.com.
7. Review and approve the design
After you submit your order, Life Technologies will email to you a BED file (UCSC Genome browser .bed
format) that contains the genomic locations targeted by the capture probes for your review and approval.
Instructions for viewing and approving the design are provided along with the BED file. For more information
on reviewing the BED file, see the TargetSeq™ Custom Enrichment Kit Ordering Guide (Part no. 4471870).
IMPORTANT! You must approve the design before Life Technologies can process your order. Once your
design is approved, manufacturing will begin and your order is irrevocable.
