German‐Israeli Foundation for Scientific Research and Development (GIF) 835/2004 Co‐investigator (€399,600 over 3 years) (PI: Prof. Dr. Rolf Hilgenfeld) The major goal of this study was to rationally design novel antibiotics targeting the bacterial stringent response by using a combination of computational chemistry, medicinal chemistry, and X‐ray crystallography. Page 4 of 21 03/06‐02/08 01/08‐12/13 01/08‐12/12 06/14‐present Intracellular feeding of Plasmodium parasites: Characterization and inhibition of the interaction between falcipain‐2 and hemoglobin. German Research Foundation (DFG) Priority Program ‘Life Inside Cells’ (SPP 1131) Co‐investigator (€105,000 over 2 years) (PI: Prof. Dr. Rolf Hilgenfeld) The major goal of this study was to elucidate the 3D structures and binding constants of the malarial protease falcipain‐2 in complex with its hemoglobin substrate to facilitate rational synthesis of novel antimalarial compounds. Autoimmunity to type VII collagen: From genetic susceptibility to novel therapeutic strategies. German Research Foundation (DFG) Excellence Cluster ‘Inflammation at Interfaces’ (EXC 306) Co‐investigator (€3.6 million over 6 years) (PI: Prof. Dr. Detlef Zillikens) The goals of this integrated and multi‐institutional study were to: (i) Elucidate susceptibility genes for blister induction by autoantibodies against type VII collagen and other cell‐matrix adhesion molecules; (ii) Determine the crystal structure of type VII collagen and its antibody‐binding sites at atomic resolution; (iii) Characterize inflammatory pathways triggered by autoantibodies resulting in loss of cell‐matrix adhesion and epithelial stem cell deletion; and (iv) Identify and therapeutically target T and B cell‐dependent mechanisms required for production of pathogenic autoantibodies against type VII collagen. *Note: I was recruited to TTUHSC‐PLFSOM prior to commencement of funding. Structural analysis of virulence‐associated proteins of Chlamydiae. German Research Foundation (DFG) Excellence Cluster ‘Inflammation at Interfaces’ (EXC 306) Principle investigator (€830,000 over 5 years) The major goal of this study was to take a structural proteomics approach to surveying the 3D structures of virulence‐associated chlamydial proteins, to use this information to predict and verify the biological function of these proteins, and to initiate structure‐based drug design strategies. *Note: I was recruited to TTUHSC‐
PLFSOM prior to commencement of funding. Structural and functional characterization of an inactive EF‐Tu mutant. TTUHSC‐PLFSOM Scholarly Activity and Research Project (SARP) Mini‐Grant Principle Investigator ($4,000 over 1 year) The focus of this study is to shed light on the structural and functional roles of conserved catalytic residues of elongation factor Tu (EF‐Tu), a key protein involved in bacterial protein biosynthesis and an important antibacterial drug target.
