Use a marker to label 7 tubes with patient’s first and last initials at the TOP of the tube and numbers 17 below the initials. 3. Place 0.3 ml of saline in each tube. 4. Place 0.3 ml of patient serum in tube 1. 5. Gently pipette up and down to mix, avoid creating air bubbles in the mixture. 6. Transfer 0.3 ml of the contents from tube 1 to tube 2. 7. Gently pipette up and down to mix, avoid creating air bubbles in the mixture. 8. Continue transferring 0.3 ml to each consecutive tube. Discard 0.3 ml from tube 7. 9. Add 0.3 ml of a 3% red blood cells solution to each tube. 10. Incubate at 4°C for 10 minutes. 11. Centrifuge for 30 seconds. 12. Gently resuspend mixture and record results below. 13. Record the interpretation of your reactions. If there is agglutination the interpretation is “positive”. If there is no agglutination, your interpretation is “negative”. 14. Calculate the dilution for each column. 15. State the titer for each column. 16. Determine the endpoint for the dilution, which is the last tube showing agglutination.
