Experimental and theoretical investigation of molecular structures of biologically active
compounds and their synthetic models, physicochemical properties of bioinorganic complexes,
pharmaceutical compounds and their models as well as food additives structures.
Experimental techniques used in these studies are mainly: Raman spectroscopy (RS),
resonance Raman spectroscopy (RR), surface enhanced (resonance) Raman spectroscopy
(SE(R)RS), time resolved resonance Raman Spectroscopy (TR 3 ), infrared spectroscopy (IR),
magnetic nuclear resonance spectroscopy (NMR), electron paramagnetic resonance
spectroscopy (EPR), mass spectrometry (MS), circular dichroism (CD) electron absorption
spectroscopy (UV-VIS). Theoretical investigations are based on ab initio and density functional
theory (DFT).

Research Proposal
Prof. L. M. Proniewicz
Small receptor-avid peptides have attracted considerable interest for use in thrombus, inflammation, and imaging/treatment of
tumors. A number of malignant tumors over-express certain types of receptors on their surface, which make them potential targets for
imaging and therapy with synthetic antagonists and agonists. Bombesin (BN) and its related peptides exhibit an enhanced response in
a variety of tumor tissues, e.g., in small cell lung, prostate, breast, and colon cancer. Therefore, the large number of modified BN
analogues and fragments have been studied, and the importance of their roles in the carcinogenesis and progression of these
diseases, have made the search for novel therapeutic ligands targeting their specific receptors a significant and growing activity in the
pharmaceutical industry. Consequently, novel approaches to evaluate the important structural components that might contribute to the
unique ability of these BN-related peptides to interact with receptors with high affinity are developed. A new approach to solve this
issue is to use modified BN analogues that are fully efficacious to the only one of BN receptors. Second methodology for the BN
screening is to apply the SERS technique for the analysis of this neurotransmitter immobilization on different metal substrates.
The use of SERS, a simple and rapid method for probing different types of supramolecular architecture and studying adsorption
phenomena at the peptide/protein level, permits to investigate the role of interfaces in hybrid electrical devices and sensors as well as
to understand structure of the adsorbed species and mechanism of binding of different substrates, i.e. neurotransmitters, to they
specific receptors. Having these information it is possible to: (1) model specific drugs, which have specific actions often targeting
hormones/neurotransmitters. For example, prozac is a serotonin reuptake inhibitor, hence potentiating its effect; AMPT prevents the
conversion of tyrosine to L-DOPA, the precursor to dopamine; reserpine prevents dopamine storage within vesicles; and deprenyl
inhibits monoamine oxidase (MAO) and thus increases dopamine levels and (2) diagnosis diseases. For example, Parkinson's disease
is at least in part related to failure of dopaminergic cells in deep-brain nuclei. The investigation effect of various mutations within the
polypeptide chain of BN is crucially important to clarify the role of each amino acid in the biological functions of these compounds. In
making synthetic or modified neuropeptides, one can sacrifice some activity in order to get a longer-lasting or more selective binding
hormone showing lower side effects.

The second aim is to establish for each investigated neuropeptide which type of amino acid modification has caused an alteration
of neuropeptide tertiary structure in turn is responsible for a marked increase/decrease in its physiological function. Then determination
of the mechanism of its biological action based on the analysis of the adsorbed structure of native form and the mutagenic analogues
and received information of they interactions (and strength) with different metal surface.
