Involvement in disease
Defects in GYS1 are the cause of muscle glycogen storage disease type 0 (GSD0b)
[MIM:611556]; also known as muscle glycogen synthase deficiency. GSD0b is a metabolic
disorder characterized by fasting hypoglycemia presenting in infancy or early childhood. The role
of muscle glycogen is to provide critical energy during bursts of activity and sustained muscle
work.

IHC image of Glycogen synthase 1 staining in
a section of formalin-fixed paraffin-embedded
normal human skeletal muscle*. The section
was pre-treated using pressure cooker heat
mediated antigen retrieval with sodium citrate
buffer (pH6) for 30mins, and incubated
overnight at +4°C with ab202546 at 1/100
dilution. DAB was used as the chromogen
(ab103723), diluted 1/100 and incubated for
10min at room temperature. The section was
counterstained with haematoxylin and

Western blot - Anti-Glycogen synthase 1 antibody
[EP817Y] (HRP) (ab202546)
Exposure time : 4 minutes
This blot was produced using a 4-12% Bistris gel under the MOPS buffer system. The
gel was run at 200V for 50 minutes before
being transferred onto a Nitrocellulose
membrane at 30V for 70 minutes. The
membrane was then blocked for an hour
using 3% milk before being incubated with
ab202546 overnight at 4°C. Antibody binding
was visualised using ECL development
solution ab133406.
